“…In the orofacial formalin test, BoNT/A injected into the whisker pad did not modulate the levels of monoamines and their metabolites, not supporting their in the BoNT/A antinociceptive action [119]. BoNT/A’s effect on ascending pain processing in humans has not been examined by functional imaging studies.…”

“…Recently, in addition to the known improvement of motor performance, an antidepressantlike effect has been demonstrated following intrastriatal injection of BoNT/A (1 ng; two injections of 1 µL solution) in a hemi-PD rat model [110]. Regarding the effects of BoNTs in depression, Ibragic et al [111] quantified the concentrations of dopamine (DA), noradrenaline (NA), serotonin (5-HT), and their metabolites in brain regions, ipsilateral, and contralateral, from the site of unilateral BoNT/A administration (5 U/Kg of Botox ® ) into the rat whisker pad. From this analysis, authors found a significant increase of NA in striatum and 5-HT in hypothalamus demonstrating an efficacy role for BoNT/A in the treatment of depression.…”

“…Increasingly, BoNT-A has been used in the treatment of chronic pain conditions like diabetic neuropathy, trigeminal neuralgia, arthritis, low back pain, myofascial pain, and has been approved for chronic migraine headache treatment in 2010. One of the most surprising new reports of toxin beneficial effect is the treatment of major depressive disorder (Ibragić S. et al, 2016) even confirmed by controlled clinical trials. In cosmetic medicine, the most widely known use of BoNT-A is the reduction of facial wrinkles, formally approved by the FDA in 2000.…”

“…The retrograde transport of catalytically active BoNT-A was found in projection neurons in cellular and animal models (Matak and Lacković, 2014). Animal data from rodents suggest the axonal transport and transcytosis in facial and spinal motor neurons, which may account for observed distant effects (Matak et al, 2016;Caleo et al, 2018;Matak, 2020). The fact that BoNT-A may undergo transcytosis and reach second order synapses suggests a process of transport very similar to tetanus neurotoxin (TeNT).…”
