Effects of bavachin and its regulation of melanin synthesis in A375 cells

Abstract: Abstract. The aim of the present study was to investigate the effect of bavachin treatment on A375 cells and the regulation of melanin synthesis. The cultured A375 cells in vitro were treated with bavachin; and the effect of bavachin on cell activity, tyrosinase (TYR) activity and melanin synthesis were respectively tested by the MTT assay, L-dopa oxidation assay and the NaOH lysis assay. The expression levels of TYR and c-Jun N-terminal kinases (JNK) proteins were tested by western blot analysis. The expressi… Show more

“…DMEM, as a medium with higher L-tyrosine content, is indicated as a factor able to induce melanization in amelanotic melanoma cells Brożyna et al 2008;Park et al 2018). There are experimental works providing for A375 culture RPMI (Scott et al 2007;Zhang and Wang 2019) or DMEM (Pal et al 2014;Wang et al 2016) media, the same referred B16F10 (Overwijk and Restifo 2000;Diawpanich et al 2008;Yoshiura et al 2009;Cunha et al 2012;Burghoff et al 2014;Potez et al 2018). Cells were harvested after 12, 24, 36, and 48 h of incubation in media, and the number of cells, melanin content, morphological changes, cell cycle, apoptosis, ROS, energetic state were analyzed.…”

“…Bavachin has been demonstrated to inhibit human hepatocellular carcinoma cells by inducing apoptosis, accompanied by ROS accumulation [ 28 ]. It has been reported that bavachin inhibits melanoma cells by downregulating the MAPK signaling pathway [ 29 ]. Notably, bavachin was shown to trigger the apoptosis of multiple myeloma cells by inhibiting p-STAT3 (705) and increasing P53 [ 30 ].…”

“…The flavonoids with prenylation group at C -6, such as bavachin, can significantly inhibit melanin synthesis and tyrosinase activity. It inhibits the expression of tyrosinase and c-Jun N-terminal kinases, and the expression of several extracellular signal-regulated kinase mRNA in A375 cells, leading to inhibition of melanin synthesis ( Wang et al, 2016 ).…”
