Background:Cyclophosphamide (CP), as an anticancer agent, causes ovarian toxicity and subsequent infertility in women. Atorvastatin (ATV) at a low dose has antioxidant and anti-inflammatory properties.Objective:The aim of this study was to investigate the protective effect of ATV against CP-induced ovarian injury in rat.Materials and Methods:In this experimental study, thirty-two female Wistar rats were randomly divided into four groups as I) control, II) ATV (10 mg/kg), III) CP (150 mg/kg), and IV) CP +ATV. The ATV treated groups were received ATV for 10 days via oral gavage. In the CP+ATV group, ATV was administrated on 5 days before and 5 days after CP injection. Histological structure, apoptosis (caspase-3), oxidative stress parameters as malondialdehyde, reactive oxygen species, protein carbonyl levels and cell viability were evaluated in ovary tissue by histological scores, immunohistochemistry, histochemical and biochemical assays. The levels of estrogen and progesterone hormones were measured on the 12th day of study.Results:ATV pretreatment significantly decreased the levels of oxidative stress biomarkers as malondialdehyde, reactive oxygen species and protein carbonyl levels and increased cell death in CP-treated rats as compared with the CP alone group. ATV significantly increased estrogen and progesterone levels in CP-treated rats. In addition, the histological examination showed ATV mitigated acute inflammation, degenerative cells in stroma and follicles, stromal edema, vacuolization, atresia of the follicles and congestion of blood vessels in the CP-treated animals. Furthermore, ATV significantly reduced immunoreactivity level of caspase-3 in CP-treated rats.Conclusion:Our results showed that the ATV with antioxidant and anti-apoptosis (caspase-3) activities protected ovarian against CP-induced toxicity.

Caffeic acid (CA) is a hydroxycinnamic acid, a group of natural organic phenolic compounds used in folk medicine and in foods for the prevention and treatment of obesity. CA is present in a variety of plants, as it is an intermediate in secondary metabolism. It is found in coffee, wine and olive oil. The aim of this work was to study the influence of caffeic acid (CA) on the reduction of intracellular lipid accumulation, intracellular reactive oxygen species (ROS) formation and in mitochondrial transmembrane potential alterations in differentiated 3T3-L1 cells. The pre-adipocyte cell line 3T3-L1 was differentiated using a differentiation cocktail. Cells were treated with CA, and the MTT assay was performed to assess the effect of CA on pre-adipocytes. The quantification of lipids accumulated within the mature cells was performed using Oil Red O dye. Flow cytometry was used to evaluate the production of ROS through 2',7'-dichlorofluorescein diacetate (DCFH-DA) oxidation and changes in mitochondrial transmembrane potential by Rhodamine 123 dye (Rho123). CA did not exhibit significant toxicity on the 3T3-L1 cell line at the studied concentrations. Caffeic acid causes significant reduction of lipid content in the cells submitted to the post-and co-treatment, being more effective in cotreatment. Caffeic acid results in an absolute decrease in the formation of intracytoplasmic reactive oxygen species. The treatment with caffeic acid protects against oxidative stress caused in the mitochondria by the adipocyte differentiation process. Thus, CA acts on adipogenesis, reducing intracellular lipid accumulation in the 3T3-L1 cells. It reduces intracellular ROS formation and mitochondrial transmembrane potential alterations in differentiated cells.

Національний фармацевтичний університет ПАТ НВЦ «Борщагівський ХФЗ»* Перспективи застосування в клінічній медицині комплексу артишоку та часникуПрепарати рослинного походження набувають у сучасному світі все більшої популярності через властиві їм пе-реваги порівняно з ліками хімічного походження. Тривалий історичний досвід застосування, високий профіль безпеки, комплексність впливу і можливість тривалого застосування без істотних побічних явищ обумовлю-ють ріст їх споживання у всьому світі. Але практика використання рослинних препаратів суттєво стримується недостатнім обсягом наукових відомостей щодо способу їх дії, потенційних побічних реакцій, протипоказань і взаємодії з іншими препаратами та їжею. В публікації розглянуті наукометричні дані щодо можливостей застосування в клінічній медицині препаратів на основі листя артишоку посівного та часнику. Проаналізована існуюча інформація, що базується на доклінічних та клінічних дослідженнях. Особлива увага приділена ре-зультатам контрольованих клінічних досліджень. Звертається увага на дуже добру переносимість та безпеку рослинних препаратів з артишоку та часнику, що зокрема дозволяє їх застосування у дітей, геронтологічних хворих, при вагітності тощо. Зроблений висновок, що згадані препарати мають значні перспективи до широ-кого застосування, а існуючі наукові дані дозволяють їх широке клінічне використання окремо та у комбінації при цілій низці захворювань та патологічних станів. At the same time, the practice of using herbal medicines is significantly hindered by the lack of scientific information about their mechanisms of action, potential adverse reactions, contraindications and interactions with other drugs and food. The publication considers scientometric data on the possible use of medicines based on leaves of artichoke and garlic in the clinical medicine. The existing information based on the preclinical and clinical studies has been analyzed. Particular attention is paid to the results of controlled clinical trials.
