Effects of antimigraine drugs on retinal spreading depression

Abstract: It has been suggested that spreading depression may play a role in triggering classical migraine. In this study the retinal spreading depression was used as a pharmacological tool to test the neuronal effects of several common antimigraine drugs. As the chicken retina is void of any blood vessels the observed effects must be of pure neuronal origin. It is shown that propranolol, sumatriptan, methysergide, paracetamol and acetylsalicyclic acid decrease the propagation velocity of retinal spreading depression wa… Show more

“…Furthermore, information could be obtained on how far the system is away from a regime where no wave propagation is possible. Recent investigations on the influence of migraine-sensitive drugs on the velocity of wave fronts (Wiedemann et al 1996) should be extended to explore their effect on two-dimensional, complex spiral wave dynamics.…”

“…Data suggest that 3-4 weeks' treatment may be necessary to achieve therapeutic efficacy, with potential for increased efficacy with longer treatment (21). Indeed, chronic administration of the drug is essential for the suppression of CSD (23)(24)(25).…”

“…Propanolol is a very effective modulator of tissue excitability reaching the maximum possible effect (50 % slowdown of spread velocity) at 400 m concentration in the bath solution [18]. Atenolol development was justified on the grounds of better affinity for the heart receptors.…”
