Effects of administration of adipose-derived stromal vascular fraction and platelet-rich plasma to dogs with osteoarthritis of the hip joints

Abstract: OBJECTIVE To evaluate effects of simultaneous intra-articular and IV injection of autologous adipose-derived stromal vascular fraction (SVF) and platelet-rich plasma (PRP) to dogs with osteoarthritis of the hip joints. ANIMALS 22 client-owned dogs (12 placebo-treated [control] dogs and 10 treated dogs). PROCEDURES Dogs with osteoarthritis of the hip joints that caused signs of lameness or discomfort were characterized on the basis of results of orthopedic examination, goniometry, lameness score, the Canine Bri… Show more

“…The surgical incision was closed routinely, and 20 g of falciform adipose tissue was processed using a commercially available kit for patient side SVF processing (ActiStem, MediVet Biologics, Nicholasville, KY) as has been described previously. 10 Briefly, preparation of the SVF involves collagenase digestion, centrifugation, and isolation of the SVF pellet. 10 The previously prepared plasma is then activated with a proprietary calcium chloride solution (Solution G, MediVet Biologics) and the SVF pellet is suspended in 2 mL of the activated plasma releasate.…”

“…[1][2][3] Stromal vascular fraction (SVF) is a point-of-care therapy that is typically prepared by the harvest of autologous adipose tissue followed by mechanical and chemical digestion and subsequent centrifugation to isolate the cellular SVF pellet from the remaining lipid and extracellular components. 10 There is no culture expansion or homogenization of the cells in the SVF so usually fewer than 5% of the cells in the SVF meet even lenient definitions of being a stem cell. [11][12][13] This is in contrast to cultured stem cell therapy in which cells are harvested from either fat or bone marrow and cultureexpanded in a laboratory over a period of weeks, a process that homogenizes the cell population while also dramatically increasing the number of available cells.…”

“…Clinical examinations were conducted 4, 8, 12, and 24 weeks after the injection. The researchers have reported a significant reduction in lameness and pain during the orthopaedic examination, and the therapeutic effect persisted for several months (42).…”

“…The best ones developed for dogs, and that have been reported to have criterion validity, are the Canine Brief Pain Inventory (CBPI) and the Liverpool Osteoarthritis in Dogs (LOAD) [9,[14][15][16]. The CBPI is divided in two sections, a pain severity score (PSS), that assesses the magnitude of the animal pain, and a pain interference score (PIS), that assesses the degree in which pain affects daily activities [17]. The Canine Orthopaedic Index (COI) was developed to assess four domains in dogs with OA: stiffness, gait, function and quality of life (QOL).…”
