Effects of Acellular Amniotic Membrane Matrix and Bone Marrow-Derived Mesenchymal Stem Cells in Improving Random Skin Flap Survival in Rats

Abstract: BackgroundThe necrotic skin flap represents a great challenge in plastic and reconstructive surgery. In this study, we evaluated the effect of bioscaffolds, acellular amniotic membranes (AAMs), and bone marrow-derived mesenchymal stem cells (BM-MSCs) on random skin flap (RSF) survival in rats by applying a cell-free extracellular matrix scaffold as a supportive component for the growth and proliferation of BM-MSCs on RSFs. AAM matrix scaffolds were created by incubating AMs in ethylenediaminetetraacetic acid 0… Show more

“…The observed limited cell growth on the surface of native amnion led us, like others [28] A variety of techniques have been used to decellularize the amnion [15], [24], [26], [28]. We optimized a trypsin-EDTA, nuclease decellularization protocol that eliminated cell and nuclear materials from the amnion.…”

“…glycosamnioglycans [17], [22], [23]. Although in most studies the amnion have been used without much modification other than dehydration, several investigations have decellularized it beforehand using a range of approaches including freeze-thawing [24], [25], EDTA [26], and trypsin-EDTA treatment [12], [13].…”

“…5 Wound healing is a complicated process that involves clot formation, inflammation, granulation, epithelialization, and remodeling. 7,8 Studies on wound healing have investigated the effect of oxygen therapy, [9][10][11] biomechanics, 12 radiotherapy and medical pomades. 13 Low-level laser therapy (LLLT) is a good modality for wound healing.…”
