Effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin on vitamin A metabolism in mice

Abstract: This study aimed to clarify the effects of single and repeated administration of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) on the activities or expression of some metabolic enzymes of retinoids and the influence of supplemental vitamin A on changed vitamin A homeostasis by TCDD. In Experiment I, the mice were given a single oral dose of 40 mug TCDD/kg body weight, with or without continuous administration of 2,500 IU vitamin A/kg body weight/day, and were killed on day 1, 3, 7, 14, and 28. In Experiment II, t… Show more

“…Interestingly ziprasidone was reported to have favorable effects on total cholesterol, LDL, and HDL levels in patients [50] , which, in context with our results, may be due to altered ABCA1 / AOX1 interaction related modulation of HDL-cholesterol levels. On the other hand, AOX1 was shown to be a dioxin inducible gene [51] . This effect may be due to AHR-mediated downregulation of PPAR [52] , as AOX1 mRNA and protein expression were recently shown do be suppressed by PPAR agonists [53] .…”

“…Residues were dissolved in 1 ml acetonitrile for each tube. Resuspended samples were analyzed immediately by HPLC using conditions as previously described ( 47 , 48 ). Briefly, HPLC was carried out on the Shimadzu LC solution system with Inertsil ® ODS-SP column (5 µ m, 4.6×150 mm) and mobile phase comprising 30 mmol/l ammonium acetate/acetonitrile (15:85, v/v).…”

“…Procedures of extracting retinoid from liver, plasma, and serum were carried out in dark room under red light to avoid retinoid degradation. Retinol and retinoic acid in liver and serum were analyzed according to previously reported method (Yang and others 2005). The linear range of retinol was 0.25 to 8 μg/mL ( r > 0.999) for the retinol in liver homogenate and 0.05 to –1 μg/mL ( r > 0.999) for the retinol in serum.…”
