Effects of 12 metal ions on iron regulatory protein 1 (IRP-1) and hypoxia-inducible factor-1 alpha (HIF-1α) and HIF-regulated genes

Abstract: Several metal ions that are carcinogenic affect cellular iron homeostasis by competing with iron transporters or iron-regulated enzymes. Some metal ions can mimic a hypoxia response in cells under normal oxygen tension, and induce expression of HIF-1α-regulated genes. This study investigated whether 12 metal ions altered iron homeostasis in human lung carcinoma A549 cells as measured by an activation of IRP-1 and ferritin level. We also studied hypoxia signaling by measuring HIF-1α protein levels, hypoxia resp… Show more

“…The depletion in ferritin levels in the Co 2 Tf-and Mn 2 Tf-treated cells is consistent with IRP activation. Indeed, Li et al (30) report IRP1 activation and reduction in ferritin protein, with Co(II) and Mn(II) treatment in A549 cells. In addition, the Co 2 Tf transport experiments suggest that transferrin can donate cobalt to the hepatoma cells, although less efficiently than it does iron, supporting the concept that cobalt and iron are both taken up by the hepatoma cells via TFRs (7).…”

“…In conditions of hypoxia this does not occur. Cobalt and manganese salts can induce chemical hypoxia via upregulation of HIF-1␣ (22,30), presumably by competing with Fe as a cofactor in this process. Similarly, iron deficiency/chelation has been shown to stabilize HIF-1␣ (2), creating conditions that mimic hypoxia.…”

“…In the literature, several authors studied the influence of oxidative stress and induced hypoxia (Antimycin, Ni and Co treatments) on iron metabolism [24][25][26][27], however they had no information about the possible change of iron oxidation state. Considering the above-mentioned experimental works and results, there is a need to follow the change of coordination and oxidative state of iron after these treatments.…”
