Effects of 1,25-dihydroxyvitamin D3 on Macrophage Cytokine Secretion Stimulated by <i>Porphyromonas gingivalis</i>

Abstract: SUMMARY: Vitamin D is known to be closely associated with periodontitis; however, its exact mechanisms remain to be clarified. The present study aimed to investigate the influence of 1,25-dihydroxyvitamin D 3 (1,25(OH) 2 D 3 ) on Porphyromonas gingivalis (Pg)-stimulated cytokine production and the involved signaling pathways in macrophages. The main observation was that 1,25(OH) 2 D 3 inhibited Pg-induced interleukin (IL)-6 cytokine expression but up-regulated the expression of antiinflammatory cytokine IL-10.… Show more

“…Research has also found that patients with low serum vitamin D levels, exhibited healing from periodontal surgery was adversely affected [100]. Related to these clinical findings, vitamin D improves epithelial innate immune functions and antimicrobial peptide secretion [101,102,103] and down-regulates NFκB activation and cytokine secretion by monocytes and macrophages [104,105]. Additionally, from the microbiome side of the periodontal disease equation, vitamin D decreases virulence gene expression by P. gingivalis [104,105].…”

“…We observed higher levels of TGF-β in vitamin D-treated EAE group. Reducing effects of vitamin D on IL-6 production have been also demonstrated in other studies (41,42). In the presence of the higher levels of TGF-β and lower levels of IL-6 (higher TGF-β/IL-6 ratio), TGF-β leads to differentiation of protective Treg cells (38).…”

“…These results indicate that calcitriol could partially abrogate aberrant host inflammatory response when exposed to gram positive bacteria. The immunomodulatory effect that calcitriol has on PAMPexposed leukocyte IL-10 production has a great deal of intra-and interspecies variability [33][34][35][36]. A previous small pilot study in dogs determined that calcitriol did not increase leukocyte production of IL-10 when exposed to LPS or LTA [11].…”
