Effectiveness of clobazam as add-on therapy in children with refractory focal epilepsy

Abstract: -The objective of this study was to evaluate the safety and efficacy of clobazam in children with refractory focal epilepsy. We investigated 100 consecutive patients concerning etiology of epilepsy, previously used antiepileptic drugs, seizure frequency and adverse events. Clobazam was introduced as add-on therapy in patients with previous failure of at least two monotherapies. Mean age was eight yearsold and 39 patients were girls. Clobazam mean dosage was 23.6 mg/day. Mean use of clobazam was 18.6 months. Tw… Show more

“…Several other studies have demonstrated the efficacy of clobazam for a variety of epilepsy syndromes and etiologies beyond Lennox-Gastaut syndrome. [6][7][8]11,[14][15][16] Eighty-five percent of the patients included in this study had a diagnosis other than Lennox-Gastaut syndrome and 9.3% were younger than 2 years of age.…”

“…4,5 Several studies have examined clobazam use in pediatric populations, with seizure freedom occurring in 9% to 60% of patients and responder rates (>50% seizure reduction) ranging from 32% to 75%. [6][7][8][9][10] Efficacy has been demonstrated when used as treatment for both localization-related and generalized epilepsy, though response of specific seizure types has only rarely been addressed.…”

“…Two retrospective studies on the efficacy of clobazam as addon therapy for pediatric patients also reported significant reductions in seizure frequency84,85 (Table 1). Da Silveira et al84 evaluated 100 patients who received clobazam as addon therapy for refractory focal epilepsy, and 33% of these patients had a 75% or greater decrease in seizure frequency.…”
