Effective three‐dimensional culture of hepatocyte‐like cells generated from human adipose‐derived mesenchymal stem cells

Abstract: Background:The aim of this study was to clarify the effectiveness of a new threedimensional (3D) culture system for hepatocyte-like cells (HLCs) generated from human adipose-derived mesenchymal stem cells (ADSCs). Methods: Human ADSCs (2 × 10 4 ) with or without 0.1 mg/mL human recombinant peptide μ-piece per well were seeded in a 96-well U-bottom plate and then our threestep differentiation protocol was applied for 21 days. At each step, cell morphology and gene expression were investigated. Mature hepatocyte… Show more

“…Therapeutic utility of mHeps has been demonstrated in a variety of different types of liver disease (Table 2). Several studies have demonstrated the engraftment of these mHeps in rodent models [19,27,28,36,[39][40][41][42][43][44][45] and pigs [46]. The efficiency of engraftment has generally not been reported, although Aurich et al presented data indicating that upwards of 20% of mouse livers were composed of mHeps at 10 weeks [41].…”

“…The RCP μ-piece acts as a scaffold for ADSCs and can support the formation of a large cell cluster. The features of this RCP include xeno-antigen-free characteristics, high cell adhesiveness, biodegradation absorption, clinical-grade quality, and stable manufacturing quality 23 . Therefore, it can achieve a cell aggregate-like technology.…”
