Effective Targeting of Colon Cancer Cells with Piperine Natural Anticancer Prodrug Using Functionalized Clusters of Hydroxyapatite Nanoparticles

Abstract: Targeted drug delivery offers great opportunities for treating cancer. Here, we developed a novel anticancer targeted delivery system for piperine (Pip), an alkaloid prodrug derived from black pepper that exhibits anticancer effects. The tailored delivery system comprises aggregated hydroxyapatite nanoparticles (HAPs) functionalized with phosphonate groups (HAP-Ps). Pip was loaded into HAPs and HAP-Ps at pH 7.2 and 9.3 to obtain nanoformulations. The nanoformulations were characterized using several techniques… Show more

“…Further coating with GoHAP resulted in new peaks centered at 1025, 601 and 586 cm −1 , corresponding to the nanoHAP powder spectrum. These peaks are associated with the phosphate stretching of P–O at 1025 cm −1 and the bending of P–O–P at 601 and 586 cm −1 [ 73 , 74 ]. By adding the drug, several peaks corresponding to it can be seen in the PEEK-HAP spectra.…”

“…Therefore, there is a need for novel formulations to improve the dissolution and solubility of PPN. A number of nanosized delivery systems have been reported for the anticancer activity of PPN, such as piperine with paclitaxel liposomes [ 15 ], apamycin and piperine polymeric nanoparticles [ 16 ], piperine micelles [ 17 ], PIP hydroxyapatite nanoparticles [ 18 ], curcumin and piperine emulsomes [ 19 ] and protopanaxadiol-piperine nanoparticles [ 20 ]. They have reported enhanced activity from individual PIP-loaded nanoformulations, as well as combined delivery with PIP with other anticancer drugs.…”

“…Piperine Emulsome combined with Curcu Emulsome (Piperine Curcumin loaded emulsomes, respectively) induced cancer cell death in HCT116 colon cancer cells in a high potential range (Bolat et al, 2020). Piperine loaded into Hydroxyapatite nanoparticles which was functionalized with phosphonate groups, successfully inhibited the monolayer of HCT116 cells and a perfect nanoformulation against colon cancer was obtained (Abouaitah et al., 2020).…”
