Effect of weak bases on the intralysosomal pH in mouse peritoneal macrophages.

Abstract: The spectral characteristics of dextran, labeled with fluorescein, depend upon pH . We have loaded the lysosomes of mouse peritoneal macrophages with this fluorescence probe and used it to measure the intralysosomal pH under various conditions . The pH of the medium has no effect on the intralysosomal pH . Weakly basic substances in the medium cause a concentration-dependent increase in the intralysosomal pH . However, the concentration of base necessary to produce a significant change in the intralysosomal pH… Show more

“…The argon laser was used to excite FITC and TMR with emission evaluated at 525 nm (pH-dependent) and 575 nm (pH-independent), respectively. Calibration of fluorescence measurements was performed using the ionophore nigericin (Sigma, St Louis, MO, USA) in buffers of known pH (Poole and Ohkuma, 1981). This allows equilibration of all the internal compartments of cells to the pH of the incubating buffer.…”

“…Here, we describe two approaches to increasing endosomal pH by coadministration of other agents that are in clinical use: (i) a competing basic compound, chloroquine (used for treatment of malaria), which enters the acidic compartments and becomes protonated, thereby raising the pH in the endosomes (Poole and Ohkuma, 1981); (ii) omeprazole (used to reduce stomach acidity), which inhibits proton pumps including that in the endosomal membrane (Mattsson et al, 1991). We also include experiments with bafilomycin A, a more specific inhibitor of the endosomal proton pump (Mattsson et al, 1991), although this compound would be too toxic to use clinically.…”

“…Chloroquine and Hydroxychloroquine have been used as autophagy inhibitors due to their action on suppression of lysosome formation and its function. 82 Chloroquine has been used in combination with many chemotherapeutic agents and a large number of trials are ongoing on its effect on cancer therapy alone or in various combinations. 83 …”

“…Ammonium chloride strongly increases the rate of protein synthesis inhibition by ITs and increases the sensitivity of cells to the IT. Raising the pH value of the lysosomes in which the ITs have been taken up is thought to be one mechanism of increasing cytotoxicity (Poole and Ohkuma, 1981). Casellas et al (1988) found that ammonium chloride could increase the cytotoxicity of ITs only when the pH was raised to above 7.…”

“…The lysosomotropic agent has been tested at 20 gg ml-', resulting in an increase in IT cytotoxicity up to 20-fold. Poole and Ohkuma (1981) have shown that weakly basic substances can increase intralysosomal pH in a concentrationdependent manner. Methylamine is a weak base that influences intralysosomal pH.…”
