“…Dinçer achieved on a MS medium containing TDZ and IBA. Our present finding is also not supported with other reports demonstrating that the urea-type cytokinins, TDZ and CPPU was more effective for the induction and the morphogenesis of adventitious multiple shoots from cotyledonary node explants of M. crystallinum plants than BAP [29].…”

“…Without a noteworthy negative side effect, BAP exerted its activity especially at higher concentrations (50-100 lM) while TDZ was already active at its lowest concentration (1 lM) but detrimental at concentrations higher than 10 lM. The higher activity towards shoot production of the ureatype cytokinin TDZ compared to adenine-type (BAP and kine) and isoprenoid-type (zea and 2iP) cytokinins, especially at lower concentrations has been reported before (Mok et al 1982;Huetteman and Preece 1993;Sunagawa et al 2007). Since we were not able to obtain somatic embryos on any of the banana shoot tips grown on different TDZ containing media, we can not confirm the property of TDZ to substitute both for auxins and cytokinins, as reported for many dicot species (Visser et al 1992;Murthy et al 1998).…”

“…The PSK has been shown to be effective in the differentiation and cell proliferation of various plant species (Igasaki et al, 2003) and to increase the transformation efficiency of carrot hypocotyl explants (Matsubayashi et al, 2004). The transformation efficiency obtained in the present study was higher than that of reports published previously on the production of transgenic ice plant (whole plant) (Sunagawa et al, 2007). It has been achieved by infection with Agrobacterium strain EHA105 harboring a binary vector pCAMBIA.…”

“…Direct organogenesis, which may possess less probability of somaclonal variation, may be more appropriate as a regeneration strategy for Agrobacterium-mediated ice plant transformation. We have developed a transformation procedure using Agrobacterium-mediated transformation of cotyledonary node explants coupled with the regeneration of transformed plants via direct organogenesis from cotyledonary node (Sunagawa, Agarie, Umemoto, Makishi & Nose, 2007). In the present study, we have examined the effects of different Agrobacterium strains and binary vectors on the efficiency of gene transfer and the effects of cytokinin, phytosulfokine (PSK) (Matsubayashi, Goto & Sakagami, 2004), NaCl and antibiotics on the efficiency of regeneration of transgenic ice plants.…”
