Acute phase proteins (APP) are biomarkers of systemic inflammation, which allow monitoring the evolution of diseases, the response to treatments, and post-operative complications. Ovariectomy (OVE) is frequently performed in veterinary medicine and can be a useful model to evaluate surgical trauma and inflammation in the bitch. The objective was to investigate and compare the acute phase response (APR) after applying three different OVE techniques by measuring serum levels of C-reactive protein (CRP), haptoglobin (Hp), albumin (Alb), and paraoxonase-1 (PON-1). Forty-five intact bitches were included in the study, being randomly distributed into three groups: laparoscopic OVE (L), midline OVE (M), and flank OVE (F). Serum CRP, Hp, Alb, and PON-1 were measured before surgery, 1, 24, 72, and 168 h post-intervention. CRP levels increased significantly 24 h post-surgery in the M and F groups, but no significant variation was observed in the L group at any time of the study period. Hp was significantly higher in group L than in group F 72 h post-surgery. Alb and PON-1 showed no statistical difference among groups or among sampling periods. CRP response suggests that the use of laparoscopic procedures produce lower inflammation compared to open conventional approaches when performing OVE in the bitch.

ZusammenfassungAufgrund von steigenden Importzahlen von Hunden aus dem Ausland, zunehmendem Reiseverkehr sowie den Veränderungen klimatischer Bedingungen in Europa gewinnen Infektionen mit Leishmania (L.) infantum bei Hunden in Deutschland zunehmend an Bedeutung. Daher sollten auch Hunde aus dem Ausland, die keine klinischen Symptome zeigen, direkt nach Import sowie erneut 6 Monate später auf vektorübertragene Infektionserreger getestet werden. Bei Hunden mit klinischer Symptomatik, die hinweisend auf eine Leishmaniose sind, werden direkte und indirekte Nachweisverfahren sowie eine hämatologische und biochemische Untersuchung unter Einbezug von Serumeiweißelektrophorese sowie Bestimmung des C-reaktiven Proteins empfohlen. Als Leitfaden für die Therapie sowie das Monitoring stehen die LeishVet-Guidelines zur Verfügung. Es stehen leishmanizide und leishmaniostatische Wirkstoffe zur Verfügung, die in first-line, second-line und third-line unterschieden werden. Zur Anpassung der Allopurinol-Dosierung wird der Stufenplan empfohlen. Aufgrund der Veränderung der klimatischen Bedingungen kommt es zu einer Ausbreitung der Habitate von Sandmücken, die als Vektoren der Leishmaniose bekannt sind. Als weitere Infektionsquellen sind Deckakte, transplazentare Infektionen, Bisswunden und Bluttransfusionen beschrieben. Leishmania infantum hat zoonotisches Potential und ist daher auch in Hinblick auf den „One-Health“-Gedanken bedeutend.

BackgroundC‐reactive protein (CRP) and paraoxonase 1 (PON1) might increase and decrease in canine leishmaniasis (CanL), , and both can rapidly normalize after therapy. Recently, supplementation of domperidone with conventional therapy , increasing the activity of cells involved in acute phase responses in vitro. This combined therapy has been recommended to treat mild forms of CanL; however, no studies have investigated the effects of domperidone supplementation on early CRP or PON1 changes in dogs with CanL.ObjectivesThe aim of this study was to evaluate whether domperidone, added to conventional treatments, modifies CRP concentration and PON1 activity kinetics in CanL dogs responsive to conventional therapy.MethodsSerum CRP concentrations and PON1 activities were measured in dogs with mild CanL before (t‐0) and 3 (t‐1), 7 (t‐2), 14 (t‐3), and 21 (t‐4) days after treatment with N‐methylglucamine antimoniate and allopurinol alone (n = 18) or combined with domperidone (n = 18).ResultsC‐reactive protein concentrations increased at t‐1 in the domperidone group, especially when the CRP concentration at t‐0 was normal. However, the concentrations normalized at t‐4 in 18/18 dogs compared with 14/18 dogs not receiving domperidone. The median PON1 activity decreased at t‐1 in the domperidone group, and this decrease was more significant in dogs with normal PON1 activity at t‐0.ConclusionsBased on these results, transient increases in CRP concentrations or decreases in PON1 activities after domperidone administration should not be erroneously interpreted as signs of a worsening disease process.
