The practice of exposing liquid cultures of the white-rot fungus Phanerochaete chrysosporium to a pure oxygen atmosphere under conditions of nutrient starvation has been widely adopted to induce lignin peroxidase (LiP) synthesis. Transmission electron microscopy was used to examine hyphal cells of carbonlimited cultures that had been exposed to an atmosphere of pure oxygen, and revealed evidence of a major loss in organization of cellular ultrastructure, which may be attributed to oxygen toxicity. Under some conditions (continuous agitation in air with cellulose as the carbon source) cultures will produce LiP without needing to be exposed to a pure oxygen atmosphere. A similar major loss of cellular ultrastructure was found in hyphal cells from such cultures upon examination. Investigation of the levels of H 2 O 2 , catalase and carbonyl content of intracellular proteins suggests that the latter cultures developed a hyperoxidant state because the rate of supply of carbon from cellulose hydrolysis was insufficient for oxygen homeostasis. The association of LiP with these cultures and with those exposed to an atmosphere of pure oxygen infers that LiP may be triggered in response to oxidant stress.

Lignin modifying enzyme (LME) activity [laccase, lignin peroxidase (LiP) and manganese peroxidase (MnP)] was studied in two indigenously isolated white rot fungi, Ganoderma species KX879638 and Lentinus species KY006984 under solid state fermentation (SSF) of wheat bran and saw dust supplemented with various non-ionic polysorbate surfactants (Tweens) and Triton X-100. Both fungal species proved to be highly ligninolytic and promising laccase producers. Their laccase activity increased many folds due to supplementation of substrates with surfactants. Among surfactants, Tween 20 was the best, giving 39180 IU/g laccase in Ganoderma spp. and 37780 IU/g in Lentinus spp. on wheat bran corresponding to 3.8-and 10.6-fold increase over control condition. Triton X-100 was the best surfactant providing 29380 and 19877 IU/g laccase activity in Ganoderma spp. and Lentinus spp. on saw dust amounting to 2.9-and 3.9-fold increase over control condition, respectively. Ganoderma spp. always produced more laccase than Lentinus spp. on supplemented substrates. Between substrates, laccase production on supplementation was always more on wheat bran than saw dust. LiP and MnP activities were very low in comparison with laccase in both fungi under control conditions (1.92 to 4.07 IU/g and 0.98 to 2.07 IU/g, respectively) and upon treatment with surfactants; they did not show an appreciable change, thus, supplementation mediated ratio between laccase to LiP/MnP was larger.

White rot fungi (WRF) are known to have the ability to degrade organic pollutants with a structure similar to lignin. Because of this, the degradation of polychlorinated biphenyls (PCBs) congeners no. 28, 52, 101, 138, 153 and 180 by substrate before fruiting (substrate) and/or after fruiting (SMS) from cultivated mushrooms Pleurotus ostreatus, Lentinula edodes and Agaricus bisporus was examined. The experiment was carried out in four replications for each treatment using a mixture of substrate/SMS and sandy soil with PCBs at a concentration of each congener at 50 and 100 µg kg -1 soil DW. The results indicate that degradation was dependent on substrate/SMS addition, the concentration of PCBs and time of incubation. The efficiency of PCB degradation was generally reduced with the number of chlorine atoms in the structure of congeners: 28, 52, 101, 138, 153 or 180. In all combinations, degradation increased with incubation time. Degradation by SMS was lower in comparison to degradation by a substrate of the same mushroom. The degree of degradation of a single PCB after 12 weeks of incubation for A. bisporus ranged from 31.32 ± 1.52 to 83.91 ± 1.07%, while for P. ostreatus it was between 37.88 ± 2.54 and 78.29 ± 1.41%; for L. edodes it ranged from 17.38 ± 1.06 to 75.30 ± 1.46%.
