Effect of TLR2 on the proliferation of inflammation-related colorectal cancer and sporadic colorectal cancer

Abstract: Background: Colitis-associated cancer (CAC) is a complication of inflammatory bowel disease (IBD) with a poor prognosis because it is often diagnosed in advanced stages with local progression or metastasis. Compared with the more common polyp-induced sporadic colorectal cancer (sCRC), CAC has different molecular mechanisms. Toll-like receptor 2 (TLR2) expression is not limited to cells related to inflammation and immune function. High levels of TLR2 expression in tumor tissues of colorectal cancer (CRC) patien… Show more

“… 323 Studies have shown that the expression level of TLR2 protein in colon cancer is significantly upregulated compared with normal epithelial tissues, 324 and the use of TLR2 agonists significantly enhances the proliferation, migration, and invasion capabilities of colon cancer cells. 325 TLR4 is highly expressed on the surface of colon cancer cells. After stimulation and activation, it can induce a variety of immunosuppressive factors, thus promoting the proliferation and immune escape of colon cancer cells.…”

“…The increased intracellular cholesterol levels eventually result in the promotion of cell proliferation [ 52 ]. As a consequence, TLR2 knockout or knockdown inhibits the proliferation of colorectal cancer cells, leading to cell cycle arrest in the G1 phase and thereby significantly impairing the development of both inflammation-related and sporadic colorectal cancer in preclinical models [ 53 ]. Of note, TLR2 is also implied in the development of chemoresistance in colorectal cancer.…”

“…The evaluation of the colonic tumors was performed according to the methods previously described [26]. The colons were cut longitudinally, the feces were removed from the colon cavity, and the colon tissue was flushed by sterile saline.…”
