Effect of thymoquinone on high fat diet and STZ‐induced experimental type 2 diabetes: A mechanistic insight by in vivo and in silico studies

Abstract: The aim was to investigate whether thymoquinone (TQ) attenuates hyperglycemia‐induced insulin resistance in experimental type 2 diabetes. Type 2 diabetes mellitus (T2DM) was induced by injection of streptozotocin (STZ, 40 mg/kg) in high fat diet (HFD) rats. The levels of glucose, insulin, area under curve (AUC) of glucose, lipid profile parameters, homeostasis model assessment of insulin resistance (HOMA‐IR), peroxisome proliferator‐activated receptor‐γ (PPARγ), and dipeptidyl peptidase peptidase‐IV (DPP‐IV) w… Show more

“…We have used the common model of rats with diabetes induced by a high-fat high-fructose (HFHF) diet and a low dose of streptozotocin (STZ) to investigate the efficacy of the phytochemical composite of Kola Joha (PCKJ) in diabetes management. 18,19 The results were confirmed through the analysis of different biochemical parameters from the collected tissues from the rats and correlated with in vitro experiments to establish the underlying mechanistic phenomena.…”

“…As for DPP-IV, the inhibitor binding cavity was utilised by TQ and surrounded by the amino acid residues Tyr547, Ser630, Tyr631, Val656, Trp659, Tyr662, Tyr666, and Val711. In addition, docked TQ upon superposition with a known inhibitor of DPP-IV displayed two robust hydrogen bond interactions with Tyr547 and Tyr662 residues [ 61 ].…”

“…TQ treatment also signi cantly improved insulin sensitivity in the treated diabetic groups. TQ prevents hyperinsulinemia, enhances the lipid pro le and insulin sensitivity, and inhibits DPP-IV to lessen the effects of hyperglycemia-related insulin resistance [1].…”
