Effect of the myeloperoxidase inhibitor AZD3241 on microglia: a PET study in Parkinson’s disease

Abstract: Impaired mitochondrial function, oxidative stress and formation of excessive levels of reactive oxygen species play a key role in neurodegeneration in Parkinson's disease. Myeloperoxidase is a reactive oxygen generating enzyme and is expressed by microglia. The novel compound AZD3241 is a selective and irreversible inhibitor of myeloperoxidase. The hypothesized mechanism of action of AZD3241 involves reduction of oxidative stress leading to reduction of sustained neuroinflammation. The purpose of this phase 2a… Show more

“…The low extent of brain exposure for gefitinib in the PET studies is consistent with human clinical experience, lending support to this approach being predictive. Indeed, PET microdosing in nonhuman primates has been used to confirm adequate brain exposure with AZD3241, a drug targeting the CNS to support to the conduct of phase IIa studies in patients (45,46).…”

“…WAPI analysis of TSPO binding has previously been shown to be sensitive to within-subject changes in V T , 37 and data based on 63-minute acquisition have shown similar reliability compared to longer timeframes. 30 To assess individual rate constants of K 1 , k 2 , k 3 , and k 4 , an additional analysis was performed using the 2-tissue compartment model (2TCM).…”
