Effect of the initial glycerol concentration in the medium on the xanthan biosynthesis

Abstract: This study is concerned with the effect of different initial glycerol concentrations in the medium on xanthan production by Xanthomonas campestris ATCC 13951. Xanthan biosynthesis was carried out in batch mode under aerobic conditions at a temperature of 30oC and agitation rate of 150 rpm for 7 days. The process efficiency was estimated based on the values of raw xanthan yield, average molecular weight of the polymer and residual content of glycerol, total nitrogen and phosphorus. Based on th… Show more

“…The highest values of this parameter were obtained in the experiments where the media with the lowest examined glycerol concentrations were used, and vice versa indicating that an increase of the carbon source content in the cultivation medium negatively affects the xanthan quality. Also, it can be noted that the values of the molecular weight are in expected ranges for such a level of biopolymer purity and in agreement with a previous study (Rončević et al 2014).…”

“…Glycerol content in cultivation media was 20.00 g/L. This concentration was selected based on the results from the previous study, conducted by the authors [22]. The cultivation medium also contained yeast extract (3.0 g/L), (NH 4 ) 2 SO 4 (1.5 g/L), K 2 HPO 4 (3.0 g/L) and MgSO 4 •7H 2 O (0.3 g/L).…”

“…The primary structure of xanthan is composed of repeating units consisting of two glucose units, two mannose units and one glucuronic acid unit in the molar ratio 2.8:2.0:2.0. Xanthan molecular weight distribution depends on the association between structural chains and it ranges from around 2•10 5 to 5•10 7 g/moL (6,7). 174 Many types of bacteria of the genus Xanthomonas such as Xanthomonas campestris, Xanthomonas phaseoli, Xanthomonas malvacearum and Xanthomonas carotae have the ability to biosynthesize xanthan, but Xanthomonas campestris is the most widely used for its production (8).…”
