Effect of the active ingredient of Kaempferia parviflora, 5,7-dimethoxyflavone, on the pharmacokinetics of midazolam

Abstract: 5,7-Dimethoxyflavone (5,7-DMF), one of the major components of Kaempferia parviflora, has anti-obesity, anti-inflammatory, and antineoplastic effects. On the other hand, in vitro studies have reported that it directly inhibits the drug metabolizing enzyme family cytochrome P450 (CYP) 3As. In this study, its safety was evaluated from a pharmacokinetic point of view, based on daily ingestion of 5,7-DMF. Midazolam, a substrate of CYP3As, was orally administered to mice treated with 5,7-DMF for 10 days, and its ph… Show more

“…The results listed by Ochiai et al [48] stated that the continued ingestion of ( 88 ) isolated from K. parviflora decreases liver CYP3A expression, which in turn increased levels of compounds metabolized by CYP3As such as midazolam [48].…”

“…This suggests that KP extract induced drug interactions mediated by CYP3A inhibition in rats in vivo . Ochiai et al reported that administration of 5,7-DMF to mice for 10 days resulted in decreased expression of hepatic CYP3A, increased AUC 0-∞ and C max of MDZ, and prolonged half-life 18 ) . In contrast, in our experiments, the effect of a single KP extract treatment was examined as a condition that does not affect expression of CYP3A, and the results obtained are considered to be mediated by CYP3A inhibition.…”

“…KP extract reportedly inhibited CYP3A activity in an in vitro experiment using mouse liver microsomes 17 ) . In addition, Ochiai et al reported that administration of 5,7-DMF to mice for ten days increased the area under the serum concentration-time curve (AUC) and elimination half-life (t 1/2 ) of midazolam (MDZ), which is a probe drug of CYP3A, and decreased hepatic CYP3A expression 18 ) . However, both reports used mice and whether the same effects will also be observed in humans is unclear.…”

“…Most of the phytochemical studies on K. parviflora in the 2020s have discussed the isolation and structural identification of methoxy flavones in rhizomes and the evaluation of its pharmaceutical properties [ 30 , 33 , 41 , 42 , 43 , 44 ]. In addition, the presence of phenolic glycosides [ 45 , 46 ], acetophenone [ 47 ], and terpenoids [ 15 , 48 , 49 , 50 ] has been reported from the rhizome extracts.…”
