Effect of textile dyes on activity and differential regulation of laccase genes from Pleurotus ostreatus grown in submerged fermentation

Abstract: This research was conducted to extend the knowledge on the differential regulation of laccase genes in response to dyes. In order to accomplish this, we analyzed both, the expression of five laccase genes by real time RT-qPCR, and also the laccase activity and isoforms patterns during the time-course of a Pleurotus ostreatus submerged fermentation supplemented with either acetyl yellow G (AYG) or remazol brilliant blue R (RBBR) dyes. For the purpose of obtaining a stable reference gene for optimal normalizatio… Show more

“…A previous study by Garrido-Bazan, et al, [10] supported the reference genes used under these conditions. The data processing of the results was initially performed using Microsoft Excel 365 and included efficiencies and reference gene normalization, while the fold expression was calculated by the 2 -Δ ΔCt method, as described by Pfaffl [11] (Eq 1).…”

“…Aside from their ecological relevance, applicability of fungi as potential biological agents in removal of industrial and agricultural effluents is getting more attention. Currently, their ability to decolorize various synthetic dyes in different reactor systems of laboratory size has been tested (Tav car et al 2006), revealing their high potential to decolorize synthetic dyes. Thus, fungi are well suited to tackle increasing problems of organic waste pollution of freshwater systems worldwide.…”

“…Laccases (EC 1.10.3.2) are multicopper oxidoreductase enzymes with the ability to oxidize a broad range of structurally differing substrates (e.g., monophenols, polyphenols, aminophenols, methoxyphenols, aromatic amines) along with the simultaneous reduction of molecular oxygen to water [1,2]. They were first described in Toxicodendron vernicifluum (Japanese lacquer tree; formerly called Rhus vernicifera) and subsequently in a wide variety of organisms, including bacteria, insects, and fungi (notably, white rot fungi) [3].…”
