Effect of testosterone and hypoxia on the expansion of umbilical cord blood CD34+ cells in vitro

Abstract: Abstract. Successfully expanding hematopoietic stem cells (HSCs) is advantageous for clinical HSC transplantation. The present study investigated the influence of testosterone on the proliferation, antigen phenotype and expression of hematopoiesis-related genes in umbilical cord blood-derived cluster of differentiation (CD)34 + cells under normoxic or hypoxia conditions. Cord blood (CB) CD34 + cells were separated using magnetic activated cell sorting. A cytokine cocktail and feeder cells were used to stimulat… Show more

“…We found that decreased erythropoiesis that results from ADTinduced profound hypogonadism is likely due to reduced stimulation of marrow erythroid precursors. Indeed, pluripotent bone marrow stem cells have androgen receptors, and stimulation by testosterone leads to differentiation of these cells toward erythroid lineage (34,50,59,70). Therefore, unliganded androgen receptor, during ADT, likely inhibits this differentiation.…”

“…During the recent decades, many evidences have been provided on the neuroendocrinology of testosterone in relation to brain activities, cognitive, emotional, and environmental behavior functions, sexual functions, mental health, suicidal behaviors, aging and pathophysiology of mood disorders, psychosis and aggression unobtrusively, and the testosterone's deficiency that may negatively affect the quality of life. [ 10 24 25 26 ] However, a few studies have evaluated the relationship between testosterone levels and bipolar disorder.…”
