IntroductionA blood test can provide important information about the functional state of the antioxidant system. Malfunction of this system increases the concentration of free radicals and can cause oxidative stress. A difficulty in assessing oxidative stress is the lack of a universal method for determining the antioxidant activity (AOA) of blood components, because of their different nature.Material and methodsThe objects of investigation were sera of 30 male patients with a diagnosis of alcohol dependence syndrome and healthy donors. Comparative investigation of total antioxidant activity (TAA) of human serum blood was carried out by voltammetric (VA), amperometric (AM) and chemiluminescent (HL) methods.ResultsAll applied methods revealed that serum TAA of the patients with alcoholism is lower than TAA of healthy donors (control group); according to amperometric method the average value of serum TAA was 850 ±210 nA × s, and 660 ±150 nA × s for healthy donors and alcoholics respectively (p < 0.05). Similar trend was revealed by chemiluminescence and voltammetry methods. The results confirm that thiol compounds make a significant contribution to the antioxidant activity of serum. The average thiol concentrations were 0.94 ±0.34 mmol/l and 1.21 ±0.36 mmol/l (p < 0.05) for alcoholics and healthy donors respectively. Decreasing thiol concentration in blood of alcoholics leads to depletion of antioxidant systems of blood. However, the differences between the results of AM, VA and HL methods were significant, because they reflected different aspects of antioxidant activity.ConclusionsFor objective assessment of antioxidant activity of biological objects, we suggest using methods based on different model systems.

This study investigates the role of oxidative stress in surgical cavernous nerve (CN) injury in a rat model. Eighty-four male Sprague-Dawley rats were randomly divided into three groups: group 1, sham-operated rats; group 2, bilateral CN-crushed rats; and group 3, bilateral CN-transection-and-sutured-immediately rats. Oxidative stress was evaluated by malondialdehyde levels, super oxide dismutase (SOD) activities, and glutathione peroxidase (GPX) activities in serum. Erectile function was assessed by CN electrostimulation at 3 months with mean maximal intracavernous pressure (ICP) and maximal ICP per mean arterial pressure. Nerve injury was assessed by toluidine blue staining of CNs and nicotinamide adenine dinucleotide phosphate (NADPH)-diaphorase staining of penile tissue. GPX protein expression and nitrotyrosine-3 (NT-3) levels in penile tissue were measured. Erectile function and the number of myelinated axons of CNs and NADPH-diaphorase-positive nerve fibers were statistically decreased between groups, from sham to crush to transection. For markers, both nerve-injury groups showed increased oxidative stress markers at early time points, with the transection group showing greater oxidative stress than the crushed group and values normalizing to sham levels by week 12. GPX expression and NT-3 levels in penile tissue were in concordance with the results of SOD and GPX. These results show that oxidative stress plays an important role in injured CNs, and different methods of CN injury can lead to different degrees of oxidative stress in a rat model.

Objective: This study evaluates the association of self-reported race with change in ankle-brachial index (ABI) over time and modification of this association by paraoxonase gene (PON1, PON2 and PON3) single nucleotide polymorphisms (SNPs). Methods: This longitudinal study included 11,992 (N = 2952 Black, N = 9040 White) participants from the Atherosclerosis Risk in Communities (ARIC) cohort with PON genotyping. Mixed-effects models examined whether race was associated with change in ABI over time after adjustment for known peripheral artery disease (PAD) risk factors. Results: Change in ABI over time differed between Whites and Blacks (race-time interaction, p < 0.0001). Stratified analyses showed that ABI values were better in both Blacks and Whites who completed high school or more education compared to those who completed less education. None of the PON SNPs met the significance level (p < 0.001) after Bonferroni correction for multiple comparisons. Conclusions: ABI differences by race were small and although statistically significant, may not be clinically significant. Change in ABI over time varies by race and may be modified by education. Results suggest that higher education may influence the lifestyle and behavioral choices contributing to better ABI in both Blacks and Whites. Further studies are needed to confirm this observation.
