Effect of Surface Morphology and Charge on the Amount and Conformation of Fibrinogen Adsorbed onto Alginate/Chitosan Microcapsules

Abstract: We report the influence of surface morphology and charge of alginate/chitosan (ACA) microcapsules on both the amount of adsorbed protein and its secondary structural changes during adsorption. Variations in surface morphology and charge were controlled by varying alginate molecular weight and chitosan concentration. Plasma fibrinogen (Fgn) was chosen to model this adsorption to foreign surfaces. The surface of ACA microcapsules exhibited a granular structure after incubating calcium alginate beads with chitosa… Show more

“…Yet, roughness is only a single measure of the topography of a surface, and despite many studies there appears to be no single unifying description of how roughness affects binding (1,2,(52)(53)(54). Our experiments are performed on surfaces with rms roughness between 1.1 nm and 0.18 nm (PD and SCD), considerably smaller than the rms roughness in most other studies (53,55).…”

“…During the membrane formation process, strong attractive forces between the nearest opposite charge groups in the polymer chains caused the contraction and aggregation of the gel skeleton as shown in Figure . The space rearrangement of gel skeleton induced the formation of big aggregates and pores on AC 1 membrane surface, which was consistent with the results observed by previous papers . However, the rougher and thicker AC 1 membrane with porous structure hardly obstructed the inward diffusion of C 2 molecules [Fig.…”

“…The surface roughness of flat membrane was measured in the wet state by a noncontact, three-dimensional optical interferometer (New-View 5020, ZYGO). 28 Before starting the measurement, free water on the surface of membranes was removed with absorbent article from one side of the gel membrane. Average values were obtained from multiple roughness values (at least three) on different areas of each sample.…”

“…As the electrolyte is introduced into the cell, the surface charges on the microcapsules are disrupted, causing a potential difference, i.e. the streaming potential (Xie et al, 2010). Electrophoresis uses an electric field to force charged particles to move in a surrounding liquid.…”
