Total oxygen consumption, glycogen content and succinic dehydrogenase levels in liver, muscle and heart in normal and thiodon-exposed (to sub-lethal concentration for 48 h and to lethal concentration for 6 h) Sarotherodon mossambicus (peters) were studied. While oxygen consumption of the fish dropped to 43 and 35% respectively under sub-lethal and lethal exposures, the magnitude of decrease in tissue glycogen content as well as tissue succinic dehydrogenase level followed a uniform order of liver> muscle> heart in both the exposures. Reduction in oxygen uptake and tissue glycogen levels were indicative of the .onset of hypoxia involving utilization of stored glycogen by the fish under thiodon exposure. The inhibition of succinic dehydrogenase levels in the 3 tissues of thiodon-exposed Sarotherodon mossambicus, while indicating the impairment of aerobic metabolism, is also suggestive of a shift from aerobic to anaerobic metabolism in the fish under pollution stress.

Abstract:The current study was designed to determine whether dietary grapeseed oil inhibits liver cytotoxicity induced by 7,12-dimethylbenz(a)anthracene (DMBA) in the frog, Rana ridibunda. The experimental animals were divided into five groups and treated for 2 weeks with 7,12-dimethylbenz(a)anthracene, DMBA plus grapeseed oil, grapeseed oil, olive oil and last group was untreated and used as control. Liver enzymes, lactate dehydrogenase, glutamic oxaloacetic acid transaminase, glutamic pyruvic acid transaminase and alkaline phosphatase were chosen to assess liver function. In comparison with control, the administration of DMBA alone significantly elevated the activity of liver lactate dehydrogenase, while the activities of glutamic oxaloacetic acid transaminase, glutamic pyruvic acid transaminase and alkaline phosphatase were declined. Similar results were noted in frogs treated with DMBA plus grapeseed oil. Moreover, it is found that the changes were more pronounced in frogs treated with DMBA plus grapeseed oil than those treated with DMBA. The activities of these enzymes in frogs exposed to grapeseed oil or olive oil were not significantly different from those of controls. These results indicate that grapeseed oil effectively increases DMBA-induced hepatotoxicity in the frogs. Also, the results suggested that grapeseed oil has enhancing effects of DMBA metabolic activation.

Chemicals present in the environment tend to alter the physiology of flora and fauna. Heavy metals tend to accumulate in the biosphere by virtue of their non-biodegradable nature. It is known that accumulation of heavy metals in tissues and organs results in cellular and molecular damage in various species (Cestari et al., 2004;Flora et al., 2012). The present study demonstrates the morphological and biochemical effects associated with the accumulation of the heavy metal lead in adult fish, Cirrhina mrigala (Common Indian Carp). Adults of Cirrhina mrigala were exposed to increasing concentrations of lead nitrate for a span of 96 hours, post which anatomical and biochemical changes were monitored. External morphology of adult fish and that of the liver and intestine were also qualitatively examined. The liver of treated fish appeared enlarged and had more mass compared with the control, whereas the intestine showed discolouration indicating ingestion of lead. Enzymes indicating stress and cellular damage were assayed in liver extracts prepared from exposed fish. The enzymes studied were succinate dehydrogenase, alanine transaminase, aspartate transaminase, acid phosphatase and alkaline phosphatase. A significant increase was foundin the activity of all of the enzymes except succinate dehydrogenase probably indicating hepatotoxicity. Succinate dehydrogenase activity was seen to have decreased, possibly indicating oxidative stress. Analysis of the neural marker enzyme acetylcholinesterase in brain extracts from these fish also showed a significant decrease in its activity which could lead to a misregulation in neurotransmission. These results directly reflect on the toxicity caused by accumulation of lead in the adult fish.Copy Right, IJAR, 2016,. All rights reserved.
