Development of chitosan based edible films: process optimization using response surface methodology

Three-factors Box-Behnken design of response surface methodology (RSM) was used to optimize chitosan level (1.5, 2.0, 2.5 %w/v), glycerol level (0.5, 0.75, 1.0 %w/v) and drying temperature (35, 40, 45°C) for the development of chitosan based edible films. The optimization was done on the basis of different responses viz. thickness, moisture, solubility, colour profile (L*, a*, b* value), penetrability, density, transmittance and water vapor transmission rate (WVTR). The linear effect of chitosan was significant (p<0.05) on all the responses. However, density was only significantly (p<0.05) affected by glycerol in a negative linear fashion. Drying temperature also significantly (p<0.05) affected thickness, penetrability, transmittance and WVTR in linear terms. The quadratic regression coefficient of chitosan showed a significant effect (p<0.05) on moisture, solubility and WVTR; glycerol level on moisture, L* value and transmittance; and drying temperature on a* value, penetrability, transmittance and WVTR. The effect of interaction of glycerol x temperature as well as chitosan x temperature was also significant (p<0.05) on a* value and WVTR of edible films. The optimized conditions were: 2.0 % w/v chitosan level, 0.75 % w/v glycerol level and drying temperature 40°C at a constant time of 48 h. All the response variables were in favourable range including thickness; 108.59 mμ, penetrability; 16.41 N, transmittance; 75.60 %, WVTR; 0.00174 g/m 2 -t for the optimized edible film. Results concluded that edible films with desirable bio-mechanical properties can be successfully developed and effectively utilized in the food packaging industry.

ABSTRAKTelah dilakukan penelitian tentang sintesis komposit film kitosan-TiO 2 menggunakan sorbitol sebagai plasticizer. Penelitian ini bertujuan untuk mengembangkan penggunaan bahan plastik yang aman bagi kesehatan dan lingkungan karena menggunakan kitosan yang biodegradabel serta TiO 2 yang tidak beracun. TiO 2 juga mampu menahan sinar UV sehingga dapat menjaga kualitas produk terhadap kerusakan akibat sinar UV. Metode penelitian dilakukan dengan sintesis TiO 2 nanokristal fasa anatase dalam matriks kitosan pada temparatur kamar dan proses aging. Larutan selanjutnya ditambahkan sorbitol dan diaduk selama 24 jam untuk menghasilkan larutan homogen. Komposit film dibentuk dengan membuat lapisan (coating) pada plat kemudian dikeringkan membentuk komposit film. Hasil karakterisasi menggunakan XRD, TEM dan FTIR menunjukkan bahwa telah terbentuk TiO 2 nanokristal fasa anatase yang terdispersi pada matriks kitosan dengan membentuk ikatan hidrogen dalam komposit film. Pada sifat fisika (physical properties) teramati bahwa penggunaan sorbitol sebagai plasticizer telah meningkatkan sifat plastis, kuat tarik (tengsile strenght) dan elongasi komposit film, tetapi pada penggunaan sorbitol yang semakin besar, sifat elastis, kuat tarik dan elongasi menjadi turun. Sifat plastis yang cukup baik dari komposit film didapatkan pada penambahan sorbitol 3 %.Kata Kunci: elongasi, kitosan-TiO 2, komposit film, kuat tarik, sorbitol. RESUMENSe estudió el efecto de factores como humedad, temperatura y tiempo de almacenamiento sobre las propiedades físicas de las películas de quitosano con y sin inclusión de aceites. Se incluyeron aceites de tomillo (TEO) y de romero (REO) en proporciones de 0,5:1 y 1:1 v/p (EOs: CH), así como su combinación 50:50 (TEO:REO). Las películas formadas se almacenaron a temperaturas de 5, 20, 33°C y humedades relativas de 60 y 93% durante cuatro semanas. Las condiciones de almacenamiento alteraron las propiedades mecánicas y físicas de las películas, tales como humedad y capacidad de retención de agua debido a la hidrofícidad del polímero, al efecto plastificante de las moléculas de agua y a fenómenos de retrogradación influenciados por la temperatura y el tiempo de almacenamiento.Palabras clave: Quitosano, polímeros, películas modificadas, aditivos alimentarios. _______________________________________________________________________________________ ABSTRACTThe effect of factors of humidity, temperature and time on the physical properties of chitosan films with and without the thyme and rosemary essential oils inclusion was studied. The thyme (TEO) and rosemary (REO) essential oils were included in 0.5:1 and 1:1 v/w (EOs:CH), as well as their combination 50:50 (TEO:REO). The films were stored at temperatures of 5, 20, 33°C and relative humidities of 60 and 93% during four weeks. Storage conditions modify the physical properties of the films due to their hydrophilic behavior, the plasticizing effect of the water molecules and the retrogradation phenomena influenced by storage temperature and time.
