Abstract: The present study was carried out to evaluate the biochemical effects of four plants extracts (D. stramonium L., Hyoscyamus muticus L., Nerium oleander L. and Eucalyptus camalulensis Dehn.) on the late instar larvae of the greater wax moth Galleria mellonella L. Galleria mellonella is used as a model organism to study the innate immune response of insects. In this study, the humoral immune response was assessed by examining phenoloxidase activity, fungal burden, and the expression of phenoloxidase and antimicrobial peptide genes at different time point following separate and combined injections of Hypericum perforatum extract and a nonlethal dose of Candida albicans. The administration of a plant extract at low doses increased phenoloxidase activity, while higher doses had no effect. Similarly, co‐injection of a low dose of the extract with the pathogen allowed half of the yeast cells to survive after 24 h. Co‐injection of plant extract with the pathogen decreased the phenoloxidase activity at the end of 4 h compared to C. albicans mono‐injection. The phenoloxidase gene expressions was reduced in all experimental conditions with respect to the control. When plant extracts and the pathogen were administered together, gallerimycin and hemolin gene expressions were considerably higher compared to mono‐injections of plant extracts and the pathogen. The results of this study reveal that gene activation and regulatory mechanisms may change for each immune gene, and that recognition and signaling pathways may differ depending on the involved immunoregulator.

The greater wax moth considered one of the most worldwide-spread pests of the bee wax. This Study was designed to investigate the insecticidal activity of Carumcarvi, Eculaptus camaldulensis and Nigella sativa against 3rd larval instar of G.mellonellathrough laboratory screening. Five concentrations (5%, 7.5%, 10%, 12.5% and 15%) of each plant extract were used in this experiment. Mortality (%) was recorded after 24, 48, 72 and 96 hrs post treatment. The results showed C. carvi, E. camaldulensis and N.sativahave insecticidal activity against 3rd_ instar larvae of G. mellonella. The highest concentration (15%) caused 90% ,86.7% and 80% larval mortality after 96 hrs for C. carvi,E. camaldulensis and N.sativarespectively. The statistical analysis revealed that there is a significant difference (p< .001) between all treatments and control. The results also clearly demonstrate that the C. carvi are significantly more toxic than E. camaldulensis and N. sativa, whereas the LC50 values were 5.4% forC. carvi,6.0% for E. camaldulensis and 6.2% for N. sativa.

The greater wax moth (Galleria mellonella) is one of the most important pests of stored or unattended combs that cause severe damage to bee broods, threatening the development of apiculture in various countries, especially Sudan. A laboratory experiment was conducted in the College of Agricultural Studies, Sudan University of Science and Technology, Shambat, Sudan, to explore the potency of ethanolic extract (EE) of different parts of three different plants and bee glue against the third larval instar of greater wax moth. The experiment was laid out in Randomized Complete Block Design and replicated five times. The percentage of mortality of GWM was recorded after 24, 48, 72, and 96 hours post-treatment. Analysis of the variance of the obtained data revealed significant differences regarding insecticidal effect between the EEs of the four test products and the control. The EE of Eucalyptus camaldulensis, Propolis, Nigella sativa, and Carum carvi have bioactivity action against the third instar larvae of GWM with an increased percentage of mortality according to the increase of concentration as well as elapse of time. Hence, the highest concentration (15%) caused 86.7%, 83.3%, 73.3 and 66.7% larval mortality after 96 hours for EE of leaves of E. camaldulensis, powder of bee glue (Propolis), seeds of N. sativa and seeds of C. carvi respectively. The results also clearly demonstrated that the EE of the powder of Propolis is significantly more toxic than E. camaldulensis, N. sativa, and C. carvi, where the LC50 values were 3.1% for Propolis, 5.0% for E. camaldulensis, 7.0 % for N. sativa and 7.7% for C. carvi. The products mentioned above at the mentioned concentration are environmental friendly, safe for honeybees, economically feasible, and affordable for small beekeepers.
