Effect of sire and extender on sperm motility and share of live or dead sperm in bulls’ fresh ejaculate and in AI doses after thawing

Abstract: The objectives were to evaluate the effects of sire and content of fresh or ionised egg yolk in extenders on sperm motility and share of live and dead sperm in collected ejaculate, in thawed artificial insemination (AI) doses, and during thermodynamic testing compared to extenders not containing egg yolk. Ejaculates were collected once a week from 4 Holstein bulls. Each of the 20 ejaculate samples from each bull was diluted with 4 different extenders. AndroMed and Bioxcell (no egg yolk) and Triladyl and Optidy… Show more

“…Collection of ejaculate and its subsequent processing present further potential risk factors for declined sperm quality. Phases of producing insemination doses (diluting of sperm, fi lling of straws, cooling and freezing) have a signifi cant eff ect on sperm motility a er thawing (Siddique et al, 2006), and the properties of extender used are especially important (Beran et al, 2012;Hegedüšová et al, 2012), as well as method of thawing (Filipčík and Hanuláková, 2011).…”

“…A phase contrast microscope with oil immersion (Eclipse E200, Nikon) at 1000× magnification was used by only one evaluator to examine all smears acquired. Minimally 100 spermatozoa were classified as either dead (with red heads) or live (with white heads), and expressed as a percentage rate of live sperm (Beran et al 2012).…”

“…As has been reported, cow conception rates may be influenced by the quality of AI doses (Beran et al, 2012) and quality of AI technicians' service (Walsh et al, 2011). In our work, only AI doses obtained from one sire were used and they were applied by one (the same) AI technician; this fact eliminates the possible influence of the sire´s individuality and service quality on the results of the performed AI.…”
