Effect of Secretin on the Secretion of Acid and Pepsin by the Totally Isolated Canine Stomach

Abstract: Totally isolated whole canine stomachs, perfused ex vivo with homologous blood, were used for study of the action of secretin on gastric secretion of acid and pepsin. During 8 h of perfusion, stomachs were stimulated by constant infusion of pentagastrin, 32 μg/h, into gastric arteries of isolated organs. When base line secretion was established (3.5 h), secretin, 60 U/h, was also infused for 90 min into the gastric artery. Secretin injection was then interrupted, but pentagastrin infusion continued until the t… Show more

“…In a previous study o f isolated canine stomachs, we used 60 U/h o f secretin infused directly into the gastric arterial system. This dose produced significant inhibition o f pentagastrin stimulated gastric secretion (19). A similar dose was used in the present experiment.…”

“…The perfusion system used in our laboratory for study of the iso lated whole canine stomach has been described previously, and illustrated in detail (10)(11)(12)(13). The stomach is dissected, immediately placed in the perfusion chamber, and connected to the perfusion circuit by way of the preceliac segment of the aorta and the distal portion of the portal vein.…”

“…We found previously that an identical dose of secretin induced a significant reduction of concentra tion and output of HC1 secretion, and a significant increase of concentration and output of pepsin secre tion, in the isolated canine stomach stimulated by pentagastrin (14). Therefore, we did not repeat the analysis of secretion for HC1 and pepsin, but limited our tests to the determination of mucinous compo nents and amino acids.…”

“…Gastric mucus secretion can be stimulated by secretin, in man (7) and in cats (8,9). Secretin is a known inhibitor of gastric acid secretion and an effective stimulant of gastric pepsin, both in vivo (13) and under ex vivo conditions (14). The effect of secretin on mucus secretion by the ex vivo perfused iso lated canine stomach is not known.…”
