Effect of Salmonella Endotoxin Administered to the Pregnant Sheep at 133–142 Days Gestation on Fetal Oxygenation, Maternal and Fetal Adrenocorticotropic Hormone and Cortisol, and Maternal Plasma Tufnor Necrosis Factor α Concentrations1

Abstract: We studied the effects of maternal exposure to endotoxin on pregnancy outcome, stimulation of the hypothalamo-hypophyseal-adrenal axes (HHAA) of ewes and their fetuses, and maternal production of tumor necrosis factor alpha (TNF alpha), a central mediator in the pathogenesis of bacterial endotoxemia. Either endotoxin (lipopolysaccharide, LPS) from Salmonella typhimurium or saline was administered to pregnant sheep (n = 10) at 137.6 +/- 3.5 days gestational age by slow i.v. infusion at 1 microgram.kg-1 total do… Show more

“…Endotoxin-induced pregnancy losses associated with embryonic death, abortion, preterm labor and delivery have been documented in a number of species (Edqvist et al 1984, Fredriksson et al 1986, Giri et al 1990, Schlafer et al 1994. However, the mechanism of LPS-induced abortion is poorly understood.…”

“…Animal models used to investigate the mechanisms whereby infection causes PTB have included the mouse, 9-12 rat, 13,14 rabbit, 15 sheep, 16 guinea pig, 17,18 and monkey. 19,20 Infectioninduced PTB can be caused by systemic or local administration of a bacterial product such as lipopolysaccharide (LPS) or endotoxin.…”

“…Clinically, it is not clear how much and by which route endotoxins enter the fetal circulation, but it is likely that the fetus is usually exposed to quite small amounts of endotoxin. In fetal sheep, doses of LPS Ͼ1 g/kg can be fatal (27), whereas doses of 1-100 g/kg are often used in studies of the adult laboratory animal (16,28). In this study, using a low dose of LPS (0.1 g/kg) that produced only transient effects on fetal pH and blood gas parameters, we found that LPS caused changes of cerebral COX-2 expression and penetration of endogenous albumin and peripheral macrophages into the brain.…”
