Effect of salivary CYP4EM1 and CYP4EM2 gene silencing on the life span of Chagas disease vector Rhodnius prolixus (Hemiptera, Reduviidae) exposed to sublethal dose of deltamethrin

Abstract: Control of Chagas disease in endemic countries is primarily accomplished through insecticide spraying for triatomine vectors. In this context, pyrethroids are the first-choice insecticide, and the evolution of insect resistance to these insecticides may represent an important barrier to triatomine control. In insects, cytochrome P450s are enzymes involved in the metabolism of xenobiotics and endogenous chemicals that are encoded by genes divided into different families. In this work, we evaluated the role of t… Show more

“…In Rhodnius prolixus , RproCYP4M1 gene is highly expressed in salivary glands (Paim et al, 2021). RNAi knockdown of RproCYP4M1 increased the susceptibility and mortality of its nymphs subsequently exposed to deltamethrin (Paim et al, 2021). As a salivary protein, CYP6P9a is associated with feeding success and blood meal size of A. funestus (Nouage et al, 2020).…”

“…Remarkably, the expression of CYP4EM1 was much higher in salivary gland than in any other organ/tissue, and it was modulated after insect blood meal. Knocking down of CYP4EM1 by RNAi induced deleterious effects in R. prolixus nymphs after topical application of deltamethrin, suggesting an important role in detoxification (29).…”

“…The role of three CYP4EM subfamily members (genes EM1, EM2 and EM3) was evaluated in vivo during R. prolixus blood meal and after deltamethrin exposure (29). A differential expression pattern of these genes was found in different insect organs and tissues; CYP4EM1 and CYP4EM2 were expressed in the integument, salivary glands, midgut, fat body and Malpighian tubules, whereas CYP4EM3 was expressed only in the integument, fat body and Malpighian tubules.…”
