Effect of Recombinant Human Bone Morphogenetic Protein-2 on Fracture Healing in a Goat Tibial Fracture Model

Abstract: Bone morphogenetic proteins (BMPs) are considered to have important regulatory roles in skeletal embryogenesis and bone healing. Recombinant human BMPs (rhBMPs) have been shown to heal critical size defects and promote spinal fusion. We studied the effects of rhBMP-2 in an absorbable collagen sponge (ACS) on bone healing in a large animal tibial fracture model. Bilateral closed tibial fractures were created in 16 skeletally mature goats and reduced and stabilized using external fixation. In each animal, one ti… Show more

“…Initially vibration upregulates expression of the osteoblast-specific master gene, Runx2, which leads to increased Osterix expression as the cells differentiate into osteoblast precursors. Runx2 and Osterix transcription factors may regulate subsequent extracellular matrix gene transcription, such as ALP, Col1, BSP, OPN, and OC (Harada et al 1999;Jang et al 2012;Javed et al 2001;Welch et al 1998). Immature osteoblasts then differentiate into mature osteoblasts through matrix production and mineralization (Eriksen 2010).…”

“…BMP-2 and BMP-7 have further been demonstrated to enhance bone healing in vivo in animals and humans in settings such as spinal fusion, fracture repair and distraction osteogenesis [23][24][25]. Although there appears to be some benefit from the use of BMPs clinically there continues to be uncertainty about the BMP potential since animal and in vitro models have demonstrated much more potent responses than results observed in cultured human cells or in clinical studies.…”

“…Bone morphogenetic proteins (BMPs), which belong to the TGF-β superfamily, have been implicated in a variety of functions including the formation of both cartilage and bone in skeletal embryogenesis and to be osteoinductive in vitro and in vivo (9). From these BMPs, BMP-2 is produced by mesenchymal cells, osteoblasts, and chondrocytes during fracture healing.…”

“…From these BMPs, BMP-2 is produced by mesenchymal cells, osteoblasts, and chondrocytes during fracture healing. BMP-2 is absolutely required for normal fracture healing (9,10). FGF-2 is recognized as a potent mitogen for a variety of mesenchymal cells.…”
