Effect of prostaglandins on luteal function during early pregnancy in pigs

Abstract: Summary. Luteal cells were obtained by digestion of luteal tissue of cyclic (day 12) and early pregnant (days 12, 20 and 30) pigs. Suspensions of the dispersed luteal cells (5 x 104 cells ml \m=-\1) were incubated for 2 h in minimum essential medium (MEM) alone (control) and MEM with different concentrations of prostaglandin FF2\ g=a\(PGF2\g=a\) and PGE2 (0\m=.\01,0\m=.\1,1, 10, 100 and 1000 ng ml \m=-\1) and luteinizing hormone (LH) 100 and 1000 ng ml\m=-\1, or with combinations of LH+ PGF2\g=a\and LH + PGE2… Show more

“…Our results showed that PGE 2 as well as PGF 2a increased secretion of P 4 by luteal slices collected from early pregnant pigs. Similar results were observed in previous studies on porcine luteal cells (Wiesak et al 1992). Moreover, PGE 2 increased P 4 synthesis and cAMP production via both PTGER2 and PTGER4.…”

“…The 30% increase of PGE 2 was non-significant in the present experiment whereas a significant increase of 300% was observed by . It is now recognized that under the influence of a conceptus signal (probably a burst of estrogen synthesis from blastocyst cells), uterine secretion of PGs changes from an endocrine direction, which exerts its luteolytic effect on corpora lutea, to an exocrine direction, which allows for continuation of the gestation (Bazer et al 1992;Wiesak et al 1992;Roberts et al 1993;Bazer et al 1994). In the present experiment, the in vitro synthesis of PGE 2 and PGF 2α by the endometrial tissue suggests that, in gilts, the efficiency of uterus tissue for PG secretion is limited and does not require large amounts of folic acid to reach its maximum potential.…”
