Effect of procarbazine on sperm morphology in syrian hamsters

Abstract: The effect of procarbazine, an antineoplastic drug, on the reproductive system of male Syrian hamsters was studied. Exposure to procarbazine (5 daily doses ranging from 20 to 500 mg/kg body weight) resulted in 5- to 7.5-fold increase in sperm abnormalities, diminished sperm counts, and smaller testes within 4 wk. Transmission electron micrographs showed severe damage to the acrosomal plasma membrane and nucleus of the sperm head in treated hamsters. These findings corroborate the detrimental effect of procarba… Show more

“…The same is true for Wistar rats, in which PCB (450 mg=kg) caused complete destruction of spermatogenesis [18]. In the hamster, exposure to PCB resulted in a 5-to 7.5-fold increase in sperm abnormalities, diminished sperm counts, and smaller testes within 4 weeks of treatment [14].…”

“…Prokarbazin uygulanan sıçanların testislerinde atrofi (151,152), Setoli ve Leydig hücrelerindeki fonksiyon bozukluğuna bağlı olarak androjen bağlayıcı protein ile testosteron düzeylerinde azalma, FSH düzeylerinde artış, ayrıca spermatogenesisteki aksaklıklar (153-155) sonucu epididimal sperm rezervlerinde azalma ve infertilite (154,155) gözlendiği bildirilmiştir. Prokarbazinle yapılan deneysel fare çalışmala-rında sperm sayısında azalma (156), hamsterlerin anormal sperm sayısında artış, sperm konsantrasyonunda azalma ile birlikte sperm başının elektron mikroskobik görüntüsünde nükleus ve akrozomal membranda şiddetli hasarlar tespit edilmiştir (157).…”
