Effect of Probenecid on the Apparent Volume of Distribution and Elimination of Cloxacillin

Abstract: According to Gibaldi et al. (1968, 1970), the higher serum concentrations of penicillins and cephaloridine reached after administration of probenecid are due not only to slower renal elimination but also to an altered distribution in the body. To determine whether probenecid has a direct effect on the distribution of cloxacillin, the elimination and distribution of cloxacillin was studied in six patients, five lacking kidney function and one with a partially impaired renal function, in the presence or absence … Show more

“…On the other hand probenecid was shown to have no affect on the distribution characteristics of cloxacillin in humans (Nauta, Mattie & Goslings, 1974), did not change the relative concentrations of ampicillin and oxacillin in fibrin clots and serum of rabbits (Barza, Brusch, Bergeron & Weinstein, 1974), and had no effect on antibacterial action of cephradine in experimentally infected mice (Kunste & Mattie, 1978).…”

“…However, the degree of active tubular transport undergone by individual ,8-lactams is highly variable (Bins & Mattie 1988). Tubular transport is blocked by probenecid (Overbosch et al 1987), as is hepatic transport (Nauta et al 1974).…”

“…Probenecid has been reported to decrease the ClI, of cloxacillin, cinoxacin, and cefazedone (13,15,18) due to competition for a common metabolic pathway or an inhibition of biliary clearance. This effect of probenecid would be most evident with drugs that are extensively metabolized.…”
