Oxytocin stimulates secretion of endometrial prostaglandin (PG) F2 alpha and induces endometrial phosphoinositide hydrolysis around the time of regression of the corpus luteum (CL) in cows. This study investigated the relationship between endometrial oxytocin receptors, oxytocin-stimulated phosphoinositide hydrolysis and PGF 2 alpha secretion in vitro, and plasma concentrations of ovarian steroids before and during CL regression in cyclic heifers (i.e., Days 13-19 post-estrus). Reproductive tracts were collected from 16 cyclic heifers on Days 13 (n = 5), 16 (n = 5), and 19 (n = 6). Decreases in mean CL weight (4.7, 4.6, and 3.0 +/- 0.6 g for Days 13, 16, and 19, respectively; p< 0.05) and plasma progesterone (12.2, 10.5, and 4.3 +/- 1.5 ng/ml for Days 13, 16, and 19, respectively; p < 0.01) were detected between Days 16 and 19, indicating that CL regression had been initiated in the group of Day 19 heifers. Mean concentration of plasma estradiol (3.4, 3.8, and 5.6 +/- 1.7 pg/ml for Days 13, 16, and 19, respectively) did not differ significantly (p > 0.5) among days of the estrous cycle. Concentration of endometrial oxytocin receptors increased (p < 0.06) during the estrous cycle (36, 49, and 789 +/- 234 fmol/mg protein on Days 13, 16, and 19, respectively), but the greatest increase (p < 0.05) occurred between Days 16 and 19. Similarly, incorporation of [3H]inositol into endometrial inositol phosphates induced in vitro with 100 nM oxytocin also increased (p < 0.01) during the estrous cycle (5121 vs. 5819, 4529 vs. 7239, and 5888 vs 68,096 +/- 18,564 dpm/g tissue for control vs. oxytocin-treated endometrium on Days 13, 16, and 19, respectively) with the greatest increase (p < 0.01) in responsiveness to oxytocin occurring between Days 16 and 19.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract. In the pig, the periimplantation period is critical for successful establishment of pregnancy. We studied the influence of embryos on oxytocin (OT) and progesterone (P4) regulated endometrial and myometrial secretion of 1) luteotrophic prostaglandin E2 (PGE2) and 2) luteolytic prostaglandin F2α and its metabolite (PGFM) on days 12-14 of pregnancy in pigs. We used unilaterally pregnant pigs created by a surgical procedure in which one uterine horn remained intact and the second horn was cut transversely so that part of the horn was detached from the uterine body. The animals were divided into two groups, inseminated gilts (days 12-14 of pregnancy, n=6) and uninseminated cyclic gilts, which were used as controls (days 12-14 of estrous cycle, n=5). Embryos developed only in the patent part of the uterus and not in the occluded horn. The abundance of OTR mRNA was increased in the endometrium and decreased in the myometrium of the gravid uterine horn in the pregnant pigs compared with the non-gravid uterine horn or either uterine horn in the cyclic pigs, indicative of a local effect of the conceptus. The presence of embryos in the uterine horn during the periimplantation period determines endometrial metabolism of PGF2α and the local response of the endometrium to OT and P4. OT stimulates PGF2α secretion and PGFM accumulation in endometrial cultures only from the non-gravid uterine horn and controls PGE2 secretion from the endometrium and myometrium in the pregnant gilts. The results indicate a more systemic affect of pregnancy on the uterine response to OT and a possibly the local effect of the conceptus in promoting progesterone's inhibition of OT-stimulated PGE2 secretion and uterine metabolism of PGF2α. Oxytocin, acting through oxytocin receptors (OTR) in the endometrium and myometrium, is involved in the control of prostaglandin F2α (PGF2α) and prostaglandin E2 (PGE2) secretion in pigs [3][4][5][6][7][8]. Prostaglandin F2α is responsible for luteal regression in non-pregnant pigs [1,9], while prostaglandin E2 acts as a luteotrophic and antiluteolytic factor that promotes maintenance of the corpus luteum during early pregnancy in pigs [10,11].Alteration of PG secretion by OT is implicated in maternal recognition of pregnancy in pigs [12][13][14]. A series of experiments were undertaken to examine the effects of interferon (IFN)-tau on regulation of prostaglandin H synthase (PGHS)-2 mRNA in bovine endometrial (BEND) cells as a means to elucidate the actions of IFN-tau to maintain pregnancy. The objective was to determine if IFN-tau mediates posttranscriptional regulation of PGHS-2 mRNA. Cells were treated with phorbol 12,13-dibutyrate (PdBu) for 3 h to induce PGHS-2 mRNA expression. Actinomycin D (0 or 1 microg/ml) or the p38 mitogen-activated protein kinase (MAPK) inhibitor, SB203580 (1 microM), were added at 3 h, followed by addition of IFN-tau (0 or 50 ng/ml) at 3.5 h and extraction of RNA at 4.5 h. The concentrations of PGHS-2 mRNA were stable between 3 and 4.5 h regardless of actinomycin D. Simultaneous treatment of PdBu-treated cells with actinomycin D and SB203580 (1 microM) decreased PGHS-2 mRNA. Addition of IFN-tau (50 ng/ml) reduced PGHS-2 mRNA, which was not observed when actinomycin D was present. Concurrent treatments of cells with SB203580 and IFN-tau (5 ng/ml) decreased concentrations of PGHS-2 mRNA in an additive manner. Although IFN-tau reduced PGHS-2 mRNA concentrations, phosphorylation of p38 MAPK was induced by IFN-tau, PdBu, and PdBu combined with IFN-tau after 10 min of treatment. Both the p38 MAPK inhibitor and IFN-tau decreased prostaglandin F(2alpha) secretion, and decreases were additive when the two were given together. In summary, activation of p38 MAPK by PdBu is required for continued presence of PGHS-2 mRNA and secretion of prostaglandin F(2alpha) in BEND cells. Interferon-tau mediates a transcription-dependent mechanism, which induces degradation of PGHS-2 mRNA. However, the consequences of an IFN-tau-induced activation of p38 MAPK warrant further investigation, because inhibition of p38 MAPK caused a degradation of PGHS-2 mRNA.
