Poverty and overpopulation are not just prevailing in big cities, but have also become a matter of concern in small towns. Safe drinking water is considered a basic human need, yet millions of people do not have access to an adequate and safe water supply. The present study is focused on the quality assessment of water bodies of slum areas of Silchar, Assam, India and to evaluate the physiological stress on Heteropneustes fossilis from household water pollution. Water samples from six different sites were collected and their physical parameters were tested. Fishes (n=35) of approximately similar size (15.8±0.84 cm) and weight (30.0±1.2 gm) were exposed to each water samples for a period of 10 days and their morphological and behavioural changes were observed. The RBC and WBC counts of the fishes were also determined. Physical properties of the water samples revealed that the pH ranges from 4.79-6.10; conductivity 0.07-0.46 mS/cm; total dissolved solids 0.03-0.29 ppm; free CO 2 13-26 ppm and dissolved oxygen 2.2-3.5 ppm. An abundance of bacteria was observed in the water samples and the predominant isolates were identified as Escherichia coli, Klebsiella sp., Bacillus sp. and Pseudomonas sp. Fishes exposed to pond water were initially active with no morphological and behavioural changes. However, from day-5 onwards, their swimming had become idle and lethargic, their dark black skin had gradually attained a pale texture and the mucus content of skin had gradually reduced. Despite providing food and air regularly, the fishes had gradually stopped eating, with swimming pattern changed from dorsal to ventral side upward and decreased opercular activity. Hematological studies showed decreased RBC count and increased WBC count in pond water treated fishes, inferring blood balance disturbance and infection in fishes. Adequate measures should be taken by the respective residents of the selected areas to minimise the degree of water contamination and use of proper disinfectants, or else the hazardous condition of the water will deteriorate the aquatic biodiversity and create serious health problems in humans.

Deterioration to extinction of wastewater bacteria by non-thermal atmospheric pressure air plasma as assessed by 16S rDNA-DGGE fingerprinting

The use of cold plasma jets for inactivation of a variety of microorganisms has recently been evaluated via culture-based methods. Accordingly, elucidation of the role of cold plasma in decontamination would be inaccurate because most microbial populations within a system remain unexplored owing to the high amount of yet uncultured bacteria. The impact of cold atmospheric plasma on the bacterial community structure of wastewater from two different industries was investigated by metagenomic-based polymerase chain reaction-denaturing gradient gel electrophoresis (DGGE) utilizing 16S rRNA genes. Three doses of atmospheric pressure dielectric barrier discharge plasma were applied to wastewater samples on different time scales. DGGE revealed that the bacterial community gradually changed and overall abundance decreased to extinction upon plasma treatment. The bacterial community in food processing wastewater contained 11 key operational taxonomic units that remained almost completely unchanged when exposed to plasma irradiation at 75.5 mA for 30 or 60 s. However, when exposure time was extended to 90 s, only Escherichia coli, Coliforms, Aeromonas sp., Vibrio sp., and Pseudomonas putida survived. Only E. coli, Aeromonas sp., Vibrio sp., and P. putida survived treatment at 81.94 mA for 90 s. Conversely, all bacterial groups were completely eliminated by treatment at 85.34 mA for either 60 or 90 s. Dominant bacterial groups in leather processing wastewater also changed greatly upon exposure to plasma at 75.5 mA for 30 or 60 s, with Enterobacter aerogenes, Klebsiella sp., Pseudomonas stutzeri, and Acidithiobacillus ferrooxidans being sensitive to and eliminated from the community. At 90 s of exposure, all groups were affected except for Pseudomonas sp. and Citrobacter freundii. The same trend was observed for treatment at 81.94 mA. The variability in bacterial community response to different plasma treatment protocols revealed that plasma had a selective impact on bacterial community structure at lower doses and potential bactericidal effects at higher doses.

Microbial community structure changes are key in detecting and characterizing the impacts of anthropogenic activities on aquatic ecosystems. Here, we evaluated the effect of river pollution of industrial and urban sites on the microbial community composition and distribution in the Nakivubo wetland and its catchment areas in Lake Victoria basin, Uganda. Samples were collected from two industrial and one urban polluted sites and the microbial diversity was analyzed based on a 16S rRNA gene clone library. Differences in microbial diversity and community structure were observed at different sampling points. Bacteria associated with bioremediation were found in sites receiving industrial waste, while a low proportion of important human-pathogenic bacteria were seen in urban polluted sites. While Escherichia spp. was the most dominant genus of bacteria for all sites, three unique bacteria, Bacillus sp., Pseudomonas sp., Thermomonas sp., which have been reported to transform contaminants such as heavy metals and hydrocarbons (such as oils) by their metabolic pathways were also identified. Our results may serve as a basis for further studies assessing microbial community structure changes among polluted sites at Nakivubo Water Channel for management and monitoring. The diversity of natural microbial consortia could also be a rich bioprospecting resource for novel industrial enzymes, medicinal and bioactive compounds.
