The present study reports on the development of a coproantigen capture enzyme-linked immunosorbent assay (ELISA) for detecting Echinostoma caproni in experimentally infected rats. The capture ELISA was based on polyclonal rabbit antibodies that recognize excretory-secretory (ES) antigens. The detection limit of pure ES was 3 ng/ml in sample buffer and 60 ng/ml in fecal samples. The test was evaluated using a follow-up of 10 rats experimentally infected with 100 metacercariae of E. caproni, and the results were compared with those of other diagnostic methods such as parasitological examination and antibody titers determined by indirect ELISA. Coproantigens were detected in all the infected rats from the first day postinfection (DPI). The period of maximal coproantigen excretion was between 7 and 21 DPI. The values remained positive until 49-56 DPI, coinciding with the disappearance of the eggs in the stool samples of the infected rats. The kinetics of coproantigen detection were correlated with those of egg output. The present assay provides an alternative tool for the diagnosis of the echinostome infections. The proposed capture ELISA makes possible an earlier diagnosis than that provided by parasitological examination and indirect ELISA and also allows for the differentiation of past and current infections. Our results show that this assay can also be used to monitor the course of echinostome infections.Echinostomes are intestinal trematodes with no tissue phase in the final host. Several vertebrates, such as aquatic birds and mammals, including humans (Graczyk and Fried, 1998), have been recorded as definitive hosts of echinostomes. Echinostomiasis in vertebrate hosts has been usually diagnosed by the presence of eggs in stool samples. However, this procedure is tedious, and eggs are not always present in feces because the worms may be preovigerous or mature worms may not be voiding eggs. Moreover, parasite maturation and egg production patterns may vary depending on different factors such as worm crowding or host species (Huffman and Fried, 1990;Toledo et al., 2003). Earlier studies have demonstrated that immunodiagnosis of echinostome infections is feasible by indirect enzyme-linked immunosorbent assay (ELISA) using different antigenic extracts. Agger et al. (1993) showed that anti-Echinostoma caproni antibodies are detectable in sera from experimentally infected NMRI mice by the 14th day postinfection (DPI) by using crude adult antigen. Graczyk and Fried (1994, 1995) detected anti-E. caproni immunoglobulins by 8 DPI in ICR mice by using glycocalyx membrane crude antigen from adult worms. However, the application of serologic diagnosis has major limitations, particularly in laboratory animals. For example, antibody titers persist after worm loss and also depend on the host species (Simonsen et al., 1991;Toledo et al., 2003). This study reports on the kinetics of antibody production to Echinostoma caproni and the dynamics of antigens in feces and sera in 2 experimental hosts (hamsters and rats) that display different degrees of susceptibility with this echinostome. Echinostoma caproni produced chronic infections in hamsters, whereas rats lost the infection at 49-56 days postinfection (DPI). Hamsters developed higher antibody responses than rats, probably in relation to different intestinal absorptions of worm antigens in each host species. The levels of coproantigens were indicative of the course of infection in each host. Positive coproantigen levels were detected at 1-2 DPI in both hosts, and the values remained positive until the end of the experiment in hamsters; in rats, the coproantigen levels reverted to negative values, coinciding with the loss of infection. High levels of circulating antigens were detected in hamsters from 21 DPI to the end of the study. In contrast, low levels of E. caproni seroantigens were detected in rats only. These observations may reflect the differences in local inflammatory responses induced by E. caproni in each host species.Infections with intestinal trematodes are widespread. Despite the frequency of these infections, the relationships between intestinal trematodes and their final hosts have received little attention. Echinostoma caproni (Trematoda: Echinostomatidae) is an intestinal trematode that does not undergo tissue migration in its definitive host. After infection of the definitive host with E. caproni, the metacercariae excyst in the duodenum, and the juvenile parasites migrate to the posterior third of the small intestine, where they attach to the mucosa by the ventral sucker (Fried and Huffman, 1996).Echinostoma caproni has a wide range of definitive hosts, although its compatibility differs considerably between rodent species. For instance, hamsters and mice show a high level of compatibility with this echinostome species, whereas rats and jirds display a low level of compatibility (Odaibo et al., , 1989Christensen et al., 1990;Hansen et al., 1991;Mahler et al., 1995; Toledo, Espert, Muñoz-Antoli et al., 2003). This classification is based mainly on worm survival observed in each host species. In highly compatible hosts E. caproni produces chronic infections, whereas in less-compatible hosts the worms are rapidly expelled. The objective of the present work was to standardize and evaluate indirect plate and dot-enzyme linked immunosorbent assay (ELISA) using purified Paramphistomum epiclitum homologous antigens in the small ruminants. Electrophoretic separation of somatic antigen (PeSAg) in reducing condition on 15% polyacrylamide gel resolved into 16 proteins of the molecular weight ranging from 14 -100 kDa. Two step ethanolic precipitation of supernatant of in-vitro culture of the fluke yielded P. epiclitum excretory-secretory antigen (PeESAg) of molecular weight 28 kDa. The animals (Goats=123; Sheep=91) were broadly kept into post-mortem and faecal examined groups. At many occasion the PeSAg found to cross reacts with other helminths parasites thus minimizing the specificity of the tests and antigens. There was no any direct correlation between the parasites load and ELISA reactivity pattern. The noted prevalence rate after combining the results of postmortem examination and PeESAg based ELISA (plate and paper/ dot) was 30.08% (37/123) in goats and 28.57% (26/91) in sheep. While using PeESAg, the calculated overall sensitivity% was 92.86 (goats)/ 100 (sheep) in both plate and dot-ELISA, specificity% was 91.58 (goats)/ 91.55 (sheep) in plate ELISA while 88.42 (goats)/ 92.96 (sheep) in dot-ELISA, positive predictive value% was 76.47 (goats)/ 76.92 (sheep) in plate ELISA while 70.27 (goats)/ 80 (sheep) in dot-ELISA and negative predictive value% was 97.75 (goats)/ 100 (sheep) in plate ELISA while 97.67 (goats)/ 100 (sheep) in dot-ELISA, these values were optimum for the field sera sample so the tests and PeESAg can be recommended for the detection P. epiclitum infection in the small ruminants.
