Effect of oxygen radical scavengers on K-cell cytolysis

Abstract: We have shown that the hydroxyl radical scavengers sodium benzoate, phenol, dimethyl sulfoxide, sodium formate, and mannitol protect chicken erythrocyte (Ec) target cells from lysis by K-cells in plaque assays. The protection afforded by benzoate, phenol, mannitol, and dimethyl sulfoxide was abrogated if the target cells were pretreated with sodium chromate. It has been shown that agents that are known to be scavengers of hydroxyl radicals may induce differentiation and inhibit growth of murine neuroblastoma cells in tissue culture. The present study tests dimethyl sulfoxide as a differentiation agent of the human neuroblastoma cell lines LA-N-1 and murine neuroblastoma NIE-115. Results indicate that DMSO induces morphologic and biochemical differentiation of neuroblastoma cells coupled to growth inhibition and inhibition of colony formation in semi liquid tissue culture systems. DMSO treatment in vitro had no effect on tumorigenicity of NIE-115 cells. In vivo DMSO treatment of athymic nude mice with transplanted LA-N-1 human neuroblastoma tumors has not affected tumor size or animal survival. No diminishing effect of natural killer cell activity could be attributed to DMSO treatment.

Borrelia species are microaerophilic spirochetes, i.e., they are unable to grow in the presence of atomospheric oxygen (21%) or under anaerobic conditions. Compounds that have been shown to quench toxic products of oxygen (i.e., superoxide radicals, hydrogen peroxide, hydroxyl radicals, and singlet oxygen) were added to Borrelia growth media (Kelly's medium). Among the compounds tested were catalase, superoxide dismutase, peroxidase, dithiothreitol (DTT), formate, fumarate, dimethyl sulfoxide (DMSO), manganese, activated charcoal (Norit A), mannitol, and a combination of ferric chloride and potassium (meta)bisulfite (iron-bisulfite). Of these, the compounds that were the most effective in enhancing the growth of the species of Borrelia tested were iron-bisulfite, DTT, and DMSO. DTT possesses the ability to break down hydrogen peroxide, and iron-bisulfite has been reported to have superoxide dismutase-like activity in artificial media. Iron-bisulfite, DMSO, sodium formate, DTT, and combinations of these compounds have been incorporated into solid and biphasic Kelley's medium to determine their effect on microaerobic and aerobic growth on plates and in biphasic medium. Iron-bisulfite was the most effective compound tested under these conditions.Key words: Borrelia, bisulfite, spirochetes, microaerobe.

To examine the possible role of reactive oxygen metabolites in lymphocyte-mediated cytolysis, the morphology of cell death following the exposure of cells to reactive oxygen metabolites in vitro was compared with the morphology of cell-mediated killing in vitro of tumour cells by natural killer (NK) cells. Ultrastructural examination of human tumour cells that were dying following incubation for 60 min with the oxygen metabolite generating systems, xanthine-xanthine oxidase or t-butylhydroperoxide, showed that cell death in both instances was exclusively by necrosis. It was unclear which oxygen metabolites were involved in killing. Cell death was not decreased by the addition of superoxide dismutase, a scavenger of the superoxide anion, to the xanthine-xanthine oxidase mixture. Although the cells were not killed by incubation with 1 mM-hydrogen peroxide, the addition of catalase, a scavenger of hydrogen peroxide, to the xanthine-xanthine oxidase mixture significantly reduced cell death. The addition of scavengers for the hydroxyl radical to either the xanthine-xanthine oxidase mixture or t-butylhydroperoxide gave inconsistent protection. In contrast, tumour cell killing mediated by natural killer cells was by apoptosis, a morphologically distinct mode of cell death with a different basic mechanism, indicating that reactive oxygen metabolites are not directly involved in lymphocyte-mediated cytolysis.
