Effect of odanacatib on root resorption and alveolar bone metabolism during orthodontic tooth movement

Abstract: ABSTRACT. The aim of this study was to investigate the effect of local administration of odanacatib (ODN) on orthodontic root resorption and the status of alveolar bone metabolism in rat molars. All specimens were scanned using microcomputed tomography and then the raw images were reconstructed. The total volume of the root resorption craters of the 60 g-NS (normal saline) group was higher than in the 60 g-ODN group and the control group. In the 60 g-NS group, the bone volume fraction values of alveolar bone w… Show more

“…Moreover, the relatively young age of the experimental dogs may have exacerbated this effect because inflammatory root resorption can occur rapidly in young teeth with wide dentinal tubules. 27 Unlike sodium fluoride, enamel matrix derivative, or bisphosphonates, which are applied once topically on the root to exert their effects, ODN may require different frequencies and modes of application for optimal results. 10,11,25 However, ODN exhibits no such characteristics and its single application before replantation may be insufficient to properly affect the healing process of the replanted tooth.…”

“…Other in vivo studies investigating the effect of ODN in the field of dentistry, although not concerned with replantation of avulsed teeth, administered ODN orally and periodically, 17,26 or injected it into the periodontium every 3 days. 27 Unlike sodium fluoride, enamel matrix derivative, or bisphosphonates, which are applied once topically on the root to exert their effects, ODN may require different frequencies and modes of application for optimal results.…”

“…It does not decrease the number of osteoclasts[ 36 ] rather decreases the magnitude of bone resorption and maintains the bone formation at the same time. [ 9 37 ] It selectively inhibits proteolysis of matrix protein by CtsK, without affecting other osteoclast activities or viability,[ 38 ] with simultaneous inhibition of inflammation. This in turn decreases osteoclast activation and differentiation, which shows a promising therapeutic method on periapical inflammatory disease.…”

“…Therefore, H(1) receptor antagonists could interfere with orthodontic treatment. 47 Another study made acomparison between three possible osteoporotic treatments in theprevention of glucocorticoidinduced alveolar bone loss. Result presented histopathologically the glucocorticoid group showed wide medullary cavities with wide osteocytic lacunae.…”
