The current study focused on testing the hypolipidemic activity of two doses of barley bran on hypercholesterolemic male rats. Twenty-four male albino rats weighing 180–200 gm were divided into four groups. The first group (G1) was the negative control, the second group (G2) was the positive control group fed 2% cholesterol in the diet, and rats of the third and the fourth groups were fed 2% cholesterol and were cosupplemented with 5% and 10% barley bran, respectively, for 8 weeks. The hypercholesterolemic rats of (G2) showed an increase in lipid profile, liver enzymes, lactate dehydrogenase, creatine kinase-MB, and lipid peroxide and a decrease in antioxidant enzymes, whereas kidney function, fasting blood sugar, glycated hemoglobin total protein, and total bilirubin were not significantly affected compared with the negative control group in G1. Moreover, histology of heart, liver, and kidney of G2 rats showed histopathological changes compared with the negative control. Administration of the two doses of barley bran in G3 and G4 to the hypercholesterolemic rats ameliorated the level of lipids, liver enzymes, lactate dehydrogenase, and creatine kinase-MB. In addition, the histology of heart, liver, and kidney tissues nearly restored the normal state as in G1.

The present study was planned to evaluate the anti-hyperlipidemic activity of theophyllin, a bronchodilator for its effect on lipid levels, HMG-CoA reductase activity and lipid peroxidation in Poloxamer 407 and high-cholesterol diet-induced hyperlipidemia in rats. The animals were divided into four groups, viz. normal, high cholesterol diet (control), Atorvastatin and theophylline treated groups. In Poloxamer-407 induced acute hyperlipidemia model, blood samples were collected at 15 and 24 hour period, and there after lipid parameters and HMG-CoA reductase activity were estimated and compared with atorvastatin reference standard treated animals. In high fat diet-induced model, animals were given respective treatment for a period of twenty one days. Lipid parameters and antioxidant parameters were measured and compared with atorvastatin treated animals at the end of study period. High cholesterol diet and Poloxamer 407 caused a significant increase in serum total cholesterol (TC), LDL-C, VLDL-C, TG and atherogenic index (AI) along with significant decrease in HDL-C and HDL/LDL ratio. In poloxamer model, Theophylline pre-treatment showed a significant decrease in total cholesterol, LDL-C, VLDL-C, and atherogenic index, with no marked change in HDL-C, TG and HDL/LDL ratio. It also showed significant inhibition of HMG-CoA reductase activity as compared to poloxamer control group. Similarly, in high fat diet model, theophylline demonstrated significant reduction in serum cholesterol, TG, VLDL and AI. In addition this, antioxidant parameters were measured in terms of Malondealdehyde levels, superoxide dismutase, catalase and reduced glutathione. Theophylline did not show any remarkable change in this parameter suggesting lack of antioxidant mechanisms. Based on our data, it is suggested that theophylline possesses hypolipidemic activity. The mechanism of action of this could be attributed to reduction (p < 0.05) in an AI and inhibition of HMG-CoA reductase activity.

Hyperlipidemia was induced in rats by administering 2% cholesterol, 20% coconut oil, and 0.125% cholic acid for 10 weeks. Atorvastatin (0.8 mg/kg b.w.) was administered orally to rats together with high-fat diet for 10 weeks. At the end of the experimental period, the circadian characteristics (acrophase, amplitude, and mesor) of liver marker enzymes (aspartate aminotransferase and alanine transferase), lipid peroxidation products (thiobarbituric acid reactive substances (TBARS), and antioxidants (superoxide dismutase, catalase, reduced glutathione, and glutathione peroxidase) were analyzed. Circadian characteristics (mesor, amplitude, and acrophase) of liver marker enzymes, TBARS, and antioxidants were altered in high-fat diet-induced rats, and the diminished amplitude along with decreased mesor levels of antioxidants were observed in high-fat diet-induced rats. Further, oral administration of atorvastatin to high-fat diet-induced rats showed the normalized mesor, amplitude, and acrophase. These findings suggest that the antihyperlipidemic potential of atorvastatin could modulate the circadian patterns of liver marker enzymes and redox status in hyperlipidemic rats.
