Effect of natural commensal-origin DNA on toll-like receptor 9 (TLR9) signaling cascade, chemokine IL-8 expression, and barrier integritiy of polarized intestinal epithelial cells

Abstract: Our study indicates that TLR9 signaling mediates, at least in part, the anti-inflammatory effects of natural commensal-origin DNA on the gut because TLR9 silencing abolished the inhibitory effect of natural commensal-origin DNA on TNF-alpha-induced IL-8 secretion in polarized IECs. The nature of the TLR9 agonist, the polarity of cells, and the tight junction integrity of IECs has to be taken into account in order to predict the outcome of TLR9 signaling. “…Examples of its proven clinical benefits include preventing and relieving certain types of diarrhea (11), reducing the incidence of respiratory infections in children (15) and impeding atopic disease (18). Up to now, only a few probiotic effector molecules, including lactic acid as an antimicrobial agent against Salmonella enterica serovar Typhimurium (7), secreted proteins that mediate homeostasis of intestinal epithelial cells (IECs) (43), and genomic DNA with anti-inflammatory effects in IECs (9), have been identified in L. rhamnosus GG. We have chosen this strain as a model for genetic studies to identify other probiotic molecules.…”

“…El epitelio intestinal está constantemente expuesto a grandes cantidades de antígenos alimenticios y bacterianos. En condiciones fisiológicas normales, la monocapa epitelial intestinal facilita un flujo controlado y selectivo de componentes entre el lúmen y la mucosa (Ghadimi et al, 2010). El balance entre la tolerancia y la inmunidad en el intestino está, en parte, determinada por poblaciones de células presentadoras de antígeno (CPA) en el intestino.…”

“…One animal study (Ohno et al, 2005) and three in vitro studies (Ghadimi et al, 2008;2010a;2010b) explored the effects of the microorganisms on the immune response to allergens/antigens. Three of these studies examined the effects of a single strain and only the in vitro …”
