Effect of nabumetone (BRL 14777), a new anti-inflammatory drug, on human platelet reactivity ex vivo: comparison with naproxen

Abstract: The effect of nabumetone (BRL 14777) on human platelet reactivity ex vivo was compared with that of naproxen at equitherapeutic doses in the same six subjects. Nabumetone had only a weak and equivocal effect on collagen-induced and second phase aggregation in response to adenosine diphosphate and adrenaline. After nabumetone, platelets fully aggregated in response to sodium arachidonate, though approximately twice as much was needed as on control occasions. Sodium arachidonate was unable to elicit a full aggre… Show more

“…In accordance with the findings of Nunn et al in healthy volunteers6 using low doses of NSAIDs, the results of our study show a significant difference in platelet aggregation, when induced with collagen 1.0 μg/ml, after the use of naproxen compared to nabumetone. Al Balla et al ,7 studying the effect of nabumetone on platelet aggregation in healthy subjects, could not demonstrate significant differences in platelet aggregation induced by higher concentrations of collagen.…”

“…Na bumetone shares many of the known side effects of NSAIDs; however, it has been sug gested that the drug has less gastrointestinal side effects than other NSAIDs [3]. Besides, the drug was also shown to have a weak anti platelet effect [4], In the present investigation we extend these earlier observations further by studying the ex vivo effects of nabumetone on the coagulation system, including platelet aggregation and bleeding time.…”

“…6-MNA competitively inhibits both cyclooxygenase isoenzymes, COX-1 and COX-2, by blocking arachidonate binding resulting in analgesic, antipyretic, and anti-inflammatory pharmacologic effects [5][6][7][8]. The enzymes COX-1 and COX-2 catalyze the conversion of arachidonic acid to prostaglandin G 2 (PGG 2 ), the first step of the synthesis of prostaglandins and thromboxanes that are generated in rapid physiological responses [9,10]. Nabumetone is used in the treatment of osteoarthritis, rheumatoid arthritis and other musculoskeletal disorders.…”
