Effect of myocyte-fibroblast coupling on the onset of pathological dynamics in a model of ventricular tissue

Abstract: Managing lethal cardiac arrhythmias is one of the biggest challenges in modern cardiology, and hence it is very important to understand the factors underlying such arrhythmias. While early afterdepolarizations (EAD) of cardiac cells is known to be one such arrhythmogenic factor, the mechanisms underlying the emergence of tissue level arrhythmias from cellular level EADs is not fully understood. Another known arrhythmogenic condition is fibrosis of cardiac tissue that occurs both due to aging and in many types … Show more

“…Fibroblasts contribute to the tissue structure, but experimental and computational studies have also shown that they can interact electrically with CMs when coupled. This results in alterations of the shape and the propagation of the action potential . A computational study by Zhan et al also suggests consequences for the mechanical contraction of CMs.…”

“…It can be viewed as an attempt to describe the transition from diffuse fibrosis to interstitial fibrosis 5 . As most previous modelling research dealt with diffuse fibrosis only [6][7][8][9][10][11] consideration of obstacle elongation makes, in our view, the next logical step toward more realistic fibrosis textures.…”

“…One such method is mathematical modelling. Most of modelling papers are devoted to diffuse fibrosis and study its effect on wave propagation and the onset of arrhythmia [6][7][8][9][10][11] . The effects of fibrosis texture on wave propagation have also been studied in a few papers.…”

“…The fact that inward current reduces both AP amplitude and repolarization rate, each of which have opposite effects on APD, can explain the mixed effects that inward currents from MFBs have been found to have on APD 80 experimentally [12,17,18,53] and computationally [49,50,[54][55][56].…”
