Effect of miR-146 targeted HDMCP up-regulation in the pathogenesis of nonalcoholic steatohepatitis

Abstract: Backgrounds/AimsMitochondrial dysfunction plays an important role inthe pathogenesis of nonalcoholic steatohepatitis (NASH), where uncoupling protein (UCP) is actively involved. We previously reported the uncoupling activity of HDMCP and its role in liver steatosis. We now aim to investigate the degree and therapeutic effect of HDMCP in NASH and the regulatory role of miR-146 on HDMCP.MethodsNASH animal model was established by feeding BALB/c mice with MCD diet while L02 cell was cultured with high concentrati… Show more

“…To gain insight into the expression changes of various miRNAs by YD during liver fibrosis, we screened several miRNAs associated with liver fibrosis and inflammation in mice by RT-PCR, including miR-125, miR-155, let-7a, miR-150, and miR-146 15 , 16 , 17 . As shown in Supporting Information Fig.…”

“…Increased expression of hepatocellular carcinoma down-regulated mitochondrial carrier protein (HDMCP) was identified in NASH animal models and HFFA-72h cultured L02 cells. The miR-146-HDMCP-downstream effector pathway is involved in NASH[ 42 ]. Collectively, previous studies have demonstrated that apoptosis resulting from mitochondrial injury is associated with the progression of NAFLD and NASH.…”

“…NASH was characterized by steatosis with inflammation. The previous study showed that upregulation of miR-146b mimic could significantly inhibit the lipid accumulation and inflammation both in vitro and in vivo [26,45]. However, despite its important roles in anti-steatosis and anti-inflammation, the potential therapeutic of miR-146b mimic has not been adequately evaluated in a translational research setting.…”
