Effect of melatonin on bovine theca cells in vitro

Abstract: Melatonin affects granulosa cell function in several species but its function in theca cells is less clear, particularly in monotocous animals. Thus, the objectives of this study were to determine the effects of melatonin on theca cell steroidogenesis, gene expression and cell proliferation in a monotocous species, namely cattle. Ovaries were collected from a local bovine abattoir, from which theca cells were isolated from large (8-22mm) follicles and treated with various hormones in serum-free medium for 24h … Show more

“…In agreement with a previous study of goat TCs, coculture with GCs did not significantly increase the expression of steroidogenic genes in TCs 20. In a previous study on bovine TCs from large follicles (8.0-22.0 mm), melatonin inhibited the LH+IGF1-induced expression of STAR mRNA 48. A possible explanation for this might be that the melatonin concentration used (10 mM = 2.3 μg/mL) was much higher than that used in the present study (10 ng/mL).…”

“…Melatonin inhibits LH + insulin-like growth factor 1 (IGF1)-induced androstenedione and progesterone production as well as the expression of steroidogenic acute regulatory protein (StAR) mRNA (via real-time polymerase chain reaction) in Theca cells. Melatonin has no effect on cytochrome P450 11A1 (CYP11A1) and cytochrome P450 17A1 (CYP17A1) mRNA abundance [ 128 ]. Melatonin increases apoptosis, induced by cisplatin, by inhibiting the JNK/Parkin/mitophagy axis [ 129 ].…”
