Effect of melatonin, curcumin, quercetin, and resveratrol on acute ferric nitrilotriacetate (Fe-NTA)-induced renal oxidative damage in rats

Abstract: The influence of melatonin, curcumin, quercetin, and resveratrol pretreatment on ferric nitrilotriacetate (Fe-NTA)-induced oxidative renal damage was studied. Male Wistar rats were treated orally once daily for 3 days with melatonin (10 mg/kg), curcumin (50 mg/kg), quercetin (15 mg/kg), and resveratrol (10 mg/kg). One hour after the last dose of antioxidants, a single dose of Fe-NTA was administered (8 mg of Fe/kg body weight, i.p.) to pre-treated animals. Twenty-four hours after Fe-NTA administration… Show more

“…Redox active transition metals have ability to induce and initiate lipid peroxidation through the production of oxygen radicals, mainly hydroxyl radical, via Fenton's/Haber-Weiss reactions [63, 406]. Transition metal, including copper [407410], chromium [411, 412], cadmium [413416], nickel [417, 418], vanadium [419421], manganese [59, 422424], and iron [59, 407, 425434] has been utilized to induce lipid peroxidation in vivo mammalian model.…”

“…Several studies have documented that smoking increases the risk of developing urinary tract cancers (20)(21)(22) and the risk for end-stage renal failure in patients with renal disease (23). Recently, several studies have shown that resveratrol exerts protective effects on acute nephrotoxicity in rats (24)(25)(26)(27) and mice (28) by suppressing the inflammatory processes and by inhibiting lipid peroxidation. Moreover, resveratrol has a positive effect on male reproductive function by triggering penile erection, as well as enhancing blood testosterone levels, testicular sperm counts, and epididymal sperm motility (29).…”

“…It ameliorates antioxidant status, reduces the expression of inflammatory biomarkers and prevents or cures cardiovascular diseases and improves microcirculatory disorders (3,4). RV, a natural antioxidant, has been suggested to reduce DNA damage and oxidative organ injury (5). Several studies demonstrated the hepatoprotective effects of RV in vivo (6,7).…”

“…Several studies demonstrated the hepatoprotective effects of RV in vivo (6,7). RV (10 mg/ kg body weight (bw)) pretreatment was found to suppress the lipid peroxidation in the kidneys of ferric nitrilotriacetate-treated rats (5). The enzyme Fpg is the major enzyme for removal of oxidized purines in bacteria and has been recommended to determine the oxidative DNA damage, in particular 8-hydroxyguanine in conjunction with the comet assay (8).…”
