Effect of medroxyprogesterone acetate (MPA) and serum factors on cell proliferation in primary cultures of an MPA-induced mammary adenocarcinoma

Abstract: The effect of progesterone (Pg), medroxyprogesterone acetate (MPA), estradiol (E2), dihydrotestosterone (DHT) and dexamethasone (DEXA) was studied on the in vitro growth rate of a progestin-dependent (PD), estrogen-sensitive mammary tumor line originated in an MPA-treated BALB/c mouse (C4-HD), and on its estrogen-resistant variant (C4-HDR). The specificity of hormone action was further investigated using the anti-hormones RU-486 and hydroxyflutamide (FLU). Cell growth was evaluated in epithelial and fibroblast… Show more

“…As we have already described, C4HD cells express high levels of PR, overexpress ErbB-2 and ErbB-3, and exhibit low ErbB-4 levels (2). Previous works have unraveled the capacity of anti-progestins to fully inhibit in vitro C4HD cell growth (13) as well as to abrogate in vivo C4HD tumor proliferation in syngeneic mice (33). We have also revealed Stat3 expression in C4HD cells (28).…”

“…Primary cultures of the C4HD murine mammary tumor line (3,4,25,44) were used. As we have already described in detail (19), C4HD epithelial cells derive from a ductal, progestin-dependent mammary tumor line. C4HD cells require MPA administration to proliferate and express high levels of PR (19).…”

“…As we have already described in detail (19), C4HD epithelial cells derive from a ductal, progestin-dependent mammary tumor line. C4HD cells require MPA administration to proliferate and express high levels of PR (19). We first investigated Stat3 expression and tyrosine phosphorylation in C4HD cells.…”

“…Primary cultures of epithelial cells from C4HD tumors, growing in MPA-treated mice, were performed as previously described (21). In brief, tumors were aseptically removed, minced, and washed with DMEM-F12 (Dulbecco modified Eagle medium-Ham F12 [1:1], without phenol red, with 100 U of penicillin/ml, and with 100 g of streptomycin/ml).…”

“…Enzyme action was stopped by adding DMEM-F12 plus 5% heat-inactivated fetal calf serum (FCS; Gen S.A., Buenos Aires, Argentina). Epithelial and fibroblastic cells were separated as previously described (21). Briefly, the cell suspension was resuspended in 15 ml of DMEM-F12 plus 10% FCS and allowed to sediment for 20 min.…”
