Effect of meal and cimetidine on endogenous plasma gastrin, secretin and pancreatic polypeptide in experimental acid hypersecretion in dogs.

Abstract: In order to observe the plasma gastrin, secretin and pancreatic polypeptide (PP) levels in an acid hypersecretion state, the pyloric antrum was transposed onto the transverse colon in 4 mongrel dogs with a Heidenhain pouch. The effects of meal and cimetidine on these gut hormones and acid secretion levels were observed for 6 hr after meals. After antrocolic transposition (ACT), hypersecretion of acid and high plasma gastrin levels were observed both in the fasting and stimulated states. Plasma secretin levels … Show more

“…Therefore, hyperinsulinemia by bombesin infusion in ACT dogs may be the combined effect of bombesin, gastrin, secretin and gastric acid on f3-cells. Postprandial hypergastrinemia, hypersecretinemia and acid hypersecretion were reported in this ACT model (Koizumi et al 1984), but it is uncertain whether chronic endogenous hypergastrinemia, hypersecretinemia and acid hypersecretion influence the fl-cell function.…”

“…Plasma gastrin, secretin and pancreatic polypeptide (PP) were measured by radioimmunoassay as previously described (Koizumi et al 1984). Plasma motilin was also measured by radioimmunoassay using the antiserum GP1103 obtained from guinea pig serum (Shin et al 1980).…”

“…Plasma was separated by centrifugation at 4°C and stored at -20©C prior to assay . Plasma immunoreactive insulin (IRI) (Morgan and Lazarow 1963), immunoreactive glucagon (IRG) (Ohneda et al 1975) and PP (Koizumi et al 1984) were measured by radioimmunoassay as described previously.…”

“…The plasma PP concentration was measured according to a previously described technique (Koizumi et al 1984). Antiserum against porcine PP (lot K-5418) was used at the final dilution of 1: 1,250,000, to bind approximately 50% of the added 1251-PPP Reference standards for the assay were prepared from PPP (lot 741029).…”
